Objective: Atrial fibrillation is one of the most common and complex clinicalarrhythmia,domestic and international study found the correlation of the cause of atrialfibrillation and the organizational structure, the electrical activity of theheart.In recentyears, the study on angiotensin-converting enzyme (ACE)polymorphism and myocardialfibrosis in atrial fibrillationhas asquired attention, wediscussed the relationship betweenthe ACE gene I/D and Kazak,han atrial fibrillation of Xinjiang in this paper To furtherclarify the molecμlar mechanisms of atrial fibrillation. Methods: Study Popμlation Thisresearch was carried out as a prospective study on400patientshospitalized in the FirstAffiliatedhospital of Xinjiang Medical Univer-sity. Among patients admitted toourhospital from January2009to August2011,100Kazakh patients andhan patientshadECG evidenced AF. The geneticdNA was extracted from200AF patients and200normalcontrols. TheI/D polymorphism of ACE wasdetected by polymerase chain reaction (PCR),and the ACE Gene I/D polymorphism of Chymase gene wasdetected using PCR restrictionfragment length polymorphism (RFLP). The genotypes and alleles of fourdifferent groupswere compared, analyses were carried out using SPSS version17.0(SPSS, Chicago, IL,USA). Thehardy-Weinberg equilibrium was assessed by chi-square analyses.differences infrequency of alleles and genotypes between studied groups were analysed using χ2test.differences were found statistically significant when P<0.05. Results: Thefrequencies ofhomozygousdeletion (DD type),heterozygous (DI type), inserthomozygotes(II type)genotype of Kazakh AF group and control group were45.0%,45.0%,10.0%and34.0%,42.0%,24.0%, The frequencies ofd allele, I allele frequency of AF group andcontrol group was67.5%,32.5%and55.0%,45.0.The frequencies ofhomozygousdeletion(DD type),heterozygous (DI type), inserthomozygotes (II type)genotype ofhan AF groupand control group were33.0%,45%,22%and18%,38%,44%, The frequencies ofd allele, I allele frequency of AF group and control group was55.5%,44.5%and37.0%,63.0%.Thefrequency of theheterozygote carriers of thedD genotype of ACE was significantlyhigherin AF patients than inhan control subjects (33%versus18%;P<0.05)andhazak subjects(45%versus34%;P<0.05). The frequency of thed allele of ACE was significantlyhigherin AF patients than inhan control subjects (55.5%versus37.0%;P<0.05)andhazaksubjects (67.5%versus55.0%;P<0.05). The frequency of thed allele of ACE wassignificantlyhigher in AF patients than inhan control subjects (34.5%versus23.0%; P<0.05)andhazak subjects (39.5%versus22.5%;P<0.05).The odds ratio (OR)for carriers ofthed allele of ACE of AF was1.699[95%confidence interval (CI):1.132~2.551] inhansubjects and2.124[95%CI:1.423~3.169] in thehazak popμlation. Conclusions: TheI/Dpolymorphism of ACE gene are associated with the occurrence of AF in Kazakh andhanEthnic AF popμlation of Xinjiang. and the genotyped might be one of the genetic riskfactors of AF. There wasdifference between Kazakh Ethnic andhan Ethnic in ACE I/Dpolymorphism allele and genotype frequency.
