The Study On Effective Substance And Evaluation Of Quality Of WXN

Thrombopenia,leucopenia and general anermia are common diseases which clinicalon the dysfunction of coagulation,hemorrhage as the main characters.TCM preparation Weixuening Heji(short as WXN,constitute with Polygoni Cuspidati Rhizoma et Radix,Spatholobi Caulis,Fried Paeoniae Radix Alba,Steamed Rehmanniae Radix,Rehmanniae Radix,Ecliptae Herba,Agrimoniae Herba and Psedostellariae Radix) is commonly used in clinical treatment of thrombocytopenia and leukopenia which has little side effect,good curative effect,good prevention,and also can be used as a tonic preparation.This paper includes review,isolation and identification,quality standard,characteristic chromatogram,fingerprint,spectral efficiency correlation,stability and preparation process.There are three parts in the paper,they are:1.The isolation and structure identification of polydatin and anthraquinone in WXN:There were4high content compound were isolated from WXN through macroporous resin and pressure silica gel column.Then identificatied them we are polydatin,emodin-8-O-beta D-glycosidase, emodin,physcion by TLC,1H-NMR,13C-NMR.2.The quality standard of WXN:（1）The medicinal materials and preparation mistura of WXN has been qualitativeidentificatied by microscope and TLC.（2）I also established three kinds of content determination methods with polydatin,emodin-8-O-beta D-glycosidase,emodin,physcion as indicators,then the quality standard of WXN was finished.（3）Characteristic chromatogram study of WXN:I have analysed ten batchs of samples of WXN by HPLC, gradient elute with acetonitrile-0.1%phosphoric acid,and evaluated them by software from SFDA.Finally,the characteristic chromatogram of WXN has been established with four chromatographic peaks,and similarity was more th an0.97.（4）Fingerprint study of WXN:I have analysed ten batchs of samples of WXN byHPLC, gradient elute with acetonitrile-0.1%phosphoric acid,and evaluated them bysoftware from SFDA.Finally,the fingerprint of WXN has been established with fivecharacteristic chromatographic peaks,and it can be used to evaluate the quality ofWXN.（5）Stability study of WXN:I established the stability and expiry date of WXN bycomparing the content of polydatin,emodin-8-O-beta D-glycosidase,emodin, physcionin the expired and unexpired WXN.3.Pharmacology and spectral efficiency correlation study of WXN:I confirmed polydatin,emodin-8-O-beta D-glycosidase were the effective compounds of WXN by Multivariable Regression model and Correlation Analysis from earlier reach conclusions of fingerprint and pharmacodynamics quantitative data.
