Objective To investigate the chemical constituents of Scabiosa tschilliensis Grunning andStudy the cytotoxic acticity of the pure compounds isolated from S. tschilliensis in vitro.Toesbatlish the quality determination method for Scabiosa tschilliensis Grunning. MethodsSolvent method was employed to extract and partition.Chemical constituents were isolatedand purified by solvent method and chromatographic method (silica gel, Sephadex LH-20,RP-18, MCI, macroporous resin HP-20, and so on).The structures of the compounds wereelucidated on the basis of physiochemical identification and spectral analysis (MS、1H-NMR、13C-NMR、DEPT and2D NMR).Cytotoxic acticity of all pure compounds were studied byMTT. Using spectrophotometric to determine the content of total flavonoids of S. tschilliensiswith reference substance of rutin. To analyze and determine the content of luteolin andapigenin in S. tschilliensis by RP-HPLC. Results sixteen compounds were isolated from S.tschilliensis and identified as oleanolic acid(1),apigenin(2),luteolin(3),daucosterol(4),kaempferol-3-O-β-D-6-O-(p-hydroxycinnamoyl)-glucopyranoside(5),kaempferol-3-O-β-D-(3,6-di-p-(hydroxycinnamoyl)-glucopyranoside(6),apigenin-7-O-β-D-glucoside(7), luteolin-4’-O-glucosid(8),apigenin-7-O-β-D-lutinoside(9),caffeic acid methyl ester(10),loganin(11),adenosine(12),luteolin-6-C-glycopyranoside(13),tschilliensisin(14),Sylvestrosides Ⅰ (15),SylvestrosidesⅡ (16),Screening test for cytotoxic acticity showed all of compounds from S.tschilliensis indicate cytotoxic effect,some of them showed anti-tumor acticity.The research ofassaying technology indicate that Samples of the total concentration of flavonoids16.448~57.568μg·mL-1within linear relationship was found.The average recovery was99.01%(RSD=1.97%).The linear range of luteolin and apigenin were7.14×10-3~42.84×10-3mg·mL-1and7.14×10-3~42.84×10-3mg·mL-1,the recovery were103.61%(RSD=0.86%) and98.82% (RSD=0.72%) respectively.Conclusions compound14is a new compound. Fourteencompounds (1、36、816) were isolated from S. tschilliensis for the frist time,twelve (36、810、1216) compounds were isolated from the genus Scabiosa for the first time.Most of thecompounds showed cytotoxic acticity.The methods esbatlished for determination of totalflavonoids,luteolin and apigenin in S. tschilliensis were simple,accurate, sensitive and can beused for assay for S. tschilliensis.
