Kusnezoff monkshood is the dried root of Ranunculaceae aconitum kusnezoffii Reichb. Leaves and flowers of Aconitum kusnezoffii Reichb also can be used as a medicine, collectively known as the Mongolian kusnezoff monkshood class. Kusnezoff monkshood has strong toxicity, clinic often associated with myrobalan, licorice compatibility to reduce their toxicity. In the research, the changes of the main ingredients in Aconitum kusnezoffii Reichb were studied to show the mechanism of detoxifying furtherly and to provide the theoretical evidence for the use of Mongolian medicine and the scientificalness of medicinal qualities.First, the contents of the main toxicity composition of double ester type alkaloids were determined to analyze the changes of the constituents before and after the compatibility of the compatibility of myrobalan and licorice of which the proportions were 1:0, 1:0.5, 1:1, 1:2, 1:3.3, 1:5, respectively. It showed that the contents of alkaloids increased first and then decreased after kusnezoff monkshood and myrobalan compatibility, which states that myrobalan joined for three kinds of double ester alkaloids content effect is remarkable, also revealed that myrobalan not only promoted the dissolution of alkaloids, but also accelerated the hydrolysis of those following the increase of myrobalan. In addition, the contents of alkaloids were reduced by mixing acomitum and licorice, which suggested that the effect of reducing the toxicity may exsit in licorice.Second, it was discovered that the contents of meaconine, hyaconitine, benzoylaconine, benzoylhyaconine and aconine decreased in the decoction after the compatibility of aconitum and myrobalan(1:2, 1:3.3, 1:5, respectively) by HPLC fingerprint. This show that as solution p H is lower and lower, accelerated the double ester alkaloids to single ester alkaloids and monkshood alkali transformation, accelerated the lower toxicity of the monoester type alkaloid and monkshood alkali hydrolysis degree at the same time, so as to achieve the role of compatibility attenuated.Finally, HPLC-MS was used to reseach the decoctions as below: aconitum decoction, myrobalan decoction and decoctions of combining aconitum and myrobalan(1:3.3, 1:5, respectively). The types and contents varied significantly according to the study. Therefore, on the basis of the standard reference sustances and reference documentions, to identify those described as follows: Fuziline(m/z 454), Karacoline(m/z 378), mesaconine(m/z 632), yunaconitine(m/z 660), aconitine(m/z 646), neoline(m/z 438), consoline(m/z 468), deoxyaconitine(m/z 630), Coke hypaconitine(m/z 556), pyraconitine(m/z 586), songorine(m/z 358), Coke deoxyaconitine(m/z 570). Then again, aconitine(m/z 646) whose content was higher and toxification was greater transfer into coke hyaconitine(m/z 556) by losing the acetoxyl on C8 and concomitantly, condensation with the ortho hydroxywhich. And coke hyaconitine(m/z 556) was transferred from hypaconitine, while following the increase of myrobalan, coke hyaconitine(m/z 556) turned into other substance losing the ionthat was [MH-CH3OH-H2O] +. In conclusion, it provided the basis for il uminating the mechanism of attenuating.
