Objective: To construct the ADAMTS-5siRNA lentivirus and investigate the effects ofADAMTS-5siRNA in cultured rat chondrocytes.Material and methods:The rat pGCSIL-GFP-ADAMTS-5siRNA plasmid wasconstructed according to the effective target sequences for RNAi of ADAMTS-5gene. TheADAMTS-5siRNA Lentivirus were produced by transfecting into packaging cell line293T. After transfected into cultured rat chondrocytes, the ADAMTS-5mRNA was testedby real-time PCR and the ADAMTDS-5protein was tested by Western blotResults: We designed and recombinated valid siRNA of rat ADAMTS-5, and packagedlentivirus vector. The transduction rate of ADAMTS-5siRNA was about80%bylentivirus vector, the expression of ADAMTS-5mRNA and ADAMTS-5protein intransducted chondrocytes was lower than the normal chondrocytes(P<0.05)Conclusions: The ADAMTS-5gene of the chondrocytes can be inhibited bylentivirus mediated siRNA, which might provide a basis for further clarify the molecularmechanism of siRNA ADAMTS-5gene silenceing in preventing and curingosteoarthritis.
