Objective:To research Cordyceps sinensis combined with Salvia miltiorrhiza organ preservation solution and observe its liver preservation effect of SD rat in vitro and vivo.Methods:(1) Developing Cordyceps sinensis combined with Salvia miltiorrhiza organ preservation solution according to the composition should be fit and tested colloid osmotic pressure, pH and electrolyte of the preservation then compared with UW solution;(2) Observing the Cordyceps sinensis combined with Salvia miltiorrhiza organ preservation solution effect through the non-establishment of SD rat liver cycle in vitro perfusion model; SD rat is randomly divided into3Group; LR liquid Group (A group), and UW solution Group (B group), and Cordyceps sinensis combined with Salvia miltiorrhiza organ preservation solution Group (C group), then under different of save time respectively will this3Group is randomly divided into save0、4、8、12、24and48hours six subgroup and wiih8SD rats in each subgroup; Respectively records bile outflow volume, and determination perfusion outflow liquid days winter aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactic acid dehydrogenation enzyme (LDH), and oxygen free base metabolism products cell aldehyde(MDA) content, determination of different PH value of the solution set of morphological changes and pathological changes of liver injury scores.(3) Establishment of SD rat liver transplantation model in vivo observation of the effect of Cordyceps sinensis combined Salvia miltiorrhiza organ preservation solution. selecting a smaller to save time on liver injury (8hours) for Orthotopic liver transplantation.The SD rat is randomly divided into Cordyceps sinensis combined with Salvia miltiorrhiza organ preservation solution group and UW solution group,wiih30animals in each group;The effects were assessed by measuring bile secretion,transplant survival rate in General, the contents of blood a week after the survival of SD rats aspartate aminotransferase (AST), alanine aminotransferase (ALT),lactate dehydrogenase (LDH) and serum bilirubin (TBIL), as well as morphological changes in liver tissue.Results:(1) Developed Cordyceps sinensis combined with Salvia miltiorrhiza organ preservation solution are tested by colloid osmotic pressure, PH and electrolyte of the preservation is no statistically significant differences compared with the UW solution.(2) The rat liver perfusion model from the body of circulation experiment, B, C group save SD rats liver bile is greater than the same period A group (P<0.01),ALT, AST, LDH value is less than the same period group A (P<0.01), B and C is no statistical difference between group (P>0.05); In the same period the MDA content B group and C group were less than A group (P<0.01), Between group C and group B is no statistical difference (P>0.05); Rat liver pathological damage rating B and C preservation group obviously less than A group in the same period(P<0.01), B and C between group was not statistically different (P>0.05); Group C prevent cell edema ia bit poor than the group B after8hours.(3) Two organ preservation solution was kept for eight hours after the SD rat liver transplantation, two groups of short-term transplant success rate was no significant difference (P>0.05); no significant difference in transplant survival after the same time bile secretion (P>0.05); one week survival rate was no significant difference (P>0.05), with the extension of survival time of bile secretion gradually increased and the color deepens, one week of bile secretion and the normal group, no significant difference (P>0.05); eating and activities gradually to normal. Survived one week whichever is the blood test for liver function suggest that no significant difference (P>0.05) between the two groups, and higher than the normal group (P<0.05), no significant difference in the morphological changes of liver tissue.Conclusion:The study indicated that the effect of self-designed Cordyceps sinensis combined with Salvia miltiorrhiza organ preservation solution on rat liver in vitio preservation are similar to that of UW solution within48hours. The bile production after reperfusion is better than its in UW group. Its the short-term survival rate and liver damage degree are no obvious difference compared with the UW solution when Save eight hours underwent orthotopic liver transplantation.
