Objective:In order to explore intervention effeets of aerobic exereise and clear-fat-herbal on the informing of high-fat-diet-indueed insulin resistance (IR) in SD rats and its mechanisms.Method:The test rats were randomly divided into5groups:Control group(C), high fat diet group (HF), high fat diet+aerobic exercise group (HE), high fat diet+clear-fat-herbal (HZ) and high fat diet+aerobic exercise+clear-fat-herbal group (HZE). The exercise plan was60min wimming without a heavy load for10weeks,6days per week. The dosage of fat-herbal filled the rats stomach was1ml/100gBW. Fasting plasma glueose(FPG),fasting plasma insulin(FINS),trigly ceride(TG),total cholesterol(TC), the blood serum tumor necrosis factor-alpha (TNF-α) and leptin were determined, at the end of the experiment, and detect the expression of mRNA of SOCS-3and SREBP-lc in the liver by real time transcription and polymeRase chain reaction (RT-PCR).Insulin sensitivity index(ISI), homeostasis model assessment-IR index (HOMA-IR), were calculated to judge the formation of IR as well as the intervention effects of aerobic exereise and clear-fat-herbal on IR.Results:1Compared with C group, blood glueose,FPG,FINS,TG,TC,FFA,leptin and TNF-a in HF group increased significantly (P<0.05,P<0.01),but IRI reduced obviously (P<0.05). By correlative analysis, FPG, FINS, TG, TC, FFA, leptin, TNF-α with HOMA-IR and ISI have significant correlation (P<0.05). Suggestion:high fat diet can induce IR by different path.2Through the double factor variance analysis,aerobic exercise supplemented clear-fat-herbal can significant reduce FPG,FINS,HOMA-IR,TG, leptin and TNF-α (P<0.05,P<0.01) and inerease ISI (P<0.05). And the aerobic exercise can reduce TC and FFA (P>0.05), but supplemented clear-fat-herbal can significant reduce them (P<0.05). Suggestion:aerobic exercise or supplemented clear-fat-herbal can improve IR induced by high fat diet in different path.3Through the double factor variance analysis, aerobic exercise united clear-fat-herbal have significant interaction in FPG, FINS, HOMA-IR, TG, TC, FFA, leptin and TNF-a (P<0.05) and inerease ISI (P<0.05). Suggestion:aerobic exercise united clear-fat-herbal have better effectiveness in improve IR induced by high fat diet than aerobic exercise or supplemented clear-fat-herbal.4Through real-time RT-PCR and double factor variance analysis, high fat diet can up-regulation the expression of mRNA of SOCS-3and SREBP-lc in the liver(P<0.01),but aerobic exercise or supplemented clear-fat-herbal can inhibit the expression (P<0.05,P<0.01),and aerobic exercise united clear-fat-herbal have preferable effect (P<0.05). By correlative analysis, FPG, FINS, HOMA-IR, TG, TC, ISI, FFA, leptin, TNF-a with SOCS-3and SREBP-lc have significant correlation (P<0.05). Suggestion:aerobic exercise united clear-fat-herbal can inhibit the expression of mRNA of SOCS-3and SREBP-lc in the liver, and regulate the level of FPG, FINS, TG, TC, FFA, leptin and TNF-a to improve IR.Conclusion:1High fat diet can improve blood lipids and increase the level of leptin and TNF-a and induce IR;2Aerobic exercise united clear-fat-herbal can improve blood lipids and decrease the level of leptin and TNF-a and inhibit IR, and aerobic exercise and clear-fat-herbal have significant correlation;3High fat diet can up-regulation the expression of mRNA of SOCS-3and SREBP-1c in the liver, but aerobic exercise and/or clear-fat-herbal can inhibit its expression, and aerobic exercise and clear-fat-herbal have significant correlation, and it is probalble mechanism of aerobic exercise and/or clear-fat-herbal inhibit IR.
