Phryma leptostachya L.var.asiatica Hara said Laopozizhenxian, Jieshengcao. It belongs to Phryma perennial herbs, and mainly distributes in hillside, forest, roadside, and grass with a great deal. It mainly distributes in the most of China with a great deal. As a medicine, it can be used with root, rhizome, stem and leaf. As a typical drug, which is concretion run smoothly, blood swelling and oxytocin, it has treated injuries, eczema, fracture, skin ulcer and so on. At present, Chinese pharmacopoeia hasn’t recorded Phryma leptostachya L.var.asiatica Hara, but it can also treat rheumatism in the folk. A comprehensive review was finished based on overall literature related to resource, the research reports of Phryma leptostachya L.var.asiatica Hara is limited, and the year of research reports is old. The study was based on the research of chemical constituents to further investigate the good method of extraction and abstraction, and else to investigate the pharmacological activity and product exploitation of this Chinese crude drug.Firstly, a comprehensive review was finished based on overall literature related to resource, chemical constituents and pharmacological activity of Phryma leptostachya L.var.asiatica Hara, which provided useful information for its further study on these aspects.Using conventional botanical chemical methods, the components of Phryma leptostachya L.var.asiatica Hara was systematically extracted. Firstly, the study which aimed on sieving the best extraction technology for Phryma leptostachya L.var.asiatica Hara. And then, the components of Phryma leptostachya L.var.asiatica Hara were systematically extracted by using petroleum ether, ethyl acetate and n-butyl alcohol. The extract of Phryma leptostachya L.var.asiatica Hara was partitioned between petroleum ether, acetic ether and n-butanol. By using GC-MS methods, the petroleum ether part was analysed of which52compounds were identified;the components which had a high content were Palmitic acid and Ethyl palmitate. Two kinds of material relative content in total28.97%. The parts of acetic ether were repeatedly subjected to the silica gel column chromatography, respectively, and4compounds were isolated from them, in which4compounds were identified as Apigenin-7-O-β-D-glucuronide, Ursolic acid, Stigmasterol, Phrymarolin-Ⅰ.So in this study, Apigenin-7-O-β-D-glucuronide as an index in the using of HPLC chromatography inspects the content of it in different parts of Phryma leptostachya L.var.asiatica Hara. Adopt the central composite design and response surface method which choose the major influential factors of extraction system. The results indicated that the highest content of Apigenin-7-O-β-D-glucuronide in leaves of Phryma leptostachya L.var.asiatica Hara. The results show that the optimum extraction process is achieve which the42.12%ethanol as extraction solvent (15.65times raw herb in volume) into materials, refluxing73.78min and extracting for3times.Using ABTS, DPPH radical and reducing reaction, determination the total extracts ethyl acetate, butanol layer and water layer extracts of Phryma leptostachya L.var.asiatica Hara in different concentrations of the antioxidant capacity. The results show that the butanol fraction from Phryma leptostachya L.var.asiatica Hara showing the most potent antioxidant activities in each assay.The study was used for determining the anti-bacterial activity of different extracts of Phryma leptostachya L.var.asiatica Hara in vitro by paper disk method. The result indicated that the butanol layer extract of Phryma leptostachya L.var.asiatica Hara had preferable anti-bacterial activity in vitro. Especially, the bacteriostatic action of staphylococcus aureus was more powerful to compare with the other strains. And else, the bacteriostatic action of salmonella was the feeblest among them.Innovative features1The components of Phryma leptostachya L have been studied. Apigenin-7-O-β-D-glucuronide was isolated for the first time.2The petroleum ether extract of Phryma leptostachya L.var.asiatica Hara has been analyzed by GC-MS for the first time, and fifty-two components were identified. The studies will provide reference for the further study of chemical constituent of Phryma leptostachya L.var.asiatica Hara.3Determination the total extracts ethyl acetate, butanol layer and water layer extracts of Phryma leptostachya L.var.asiatica Hara in different concentrations of the antioxidant capacity for the first time. The result indicated that the butanol fraction from Phryma leptostachya L.var.asiatica Hara showing the most potent antioxidant activities in each assay.4The study preliminarily investigated vitro-antibacterial activity of different extracts of Phryma leptostachya L.var.asiatica Hara for the first time. The result indicated that the butanol layer extract of Phryma leptostachya L.var.asiatica Hara has preferable antibacterial action in vitro, especially the bacteriostatic action of staphylococcus aureus was the most powerful.
