Objective:Discussing the changes of blood hemorheology of early fatty liver in rat and therole of Quzhi Ruangan Decoction in hemorheological intervention, confirming therelationship of early fatty liver, hemorheological and blood stasis syndrome oftraditional Chinese medicine, which will provide laboratory basis for the clinicalusage of Quzhi Ruangan Decoction, treating early fatty liver.Methods:Ⅰ. Preparations:24healthy and male rats are randomized into a normal group, amodel group and a Quzhi Ruangan Decoction group, each group has8rats and is fed8weeks respectively. The normal group is fed with normal animal forages, while theother two groups are fed with high fat forage (adding10%of lard oil and2%ofcholestenone to normal animal forages). The rats of each group can drink water freely.Ⅱ. Feeding medicine to each group: the rats of normal group and model groupwere given a gavage of1ml physiological saline per day; the rats of Quzhi RuanganDecoction group were given a gavage of1ml Quzhi Ruangan Decoction soup per day,which was calculated by clinical equivalent amount. The rats in each group will befed8weeks respectively.Ⅲ.Index detecting: modeling and intervening treatment on rats to the end of the8th week, taking the blood sample and liver tissue sample from the rats in each grouprespectively, testing the weight and the liver wet weight of rat; calculating the rat’sliver weigh index to weight; observing the liver fatty degeneration by hematoxylin and eosin(HE) staining; testing the rat’s blood serum and liver function by alanineaminotransferase and aspartateaminotransferase(AST); testing rat’s cholesterof(TC)and triaglyceride(TG); testing the liver lipid; testing the hemorheology of whole bloodviscosity, plasma viscosity, aggregative index of red blood cell, fibrinogen, packedcell volume and platelet aggregation rate.Results:Ⅰ. The models of early fatty liver in rats by feeding rats with high fatty forage.The rats in each group are in good condition before experiment. The rats of modelgroup are becoming fat from the fourth week and the rats showing the condition of furgloss reducing, food-intake decreasing, and mobility reducing and sticky, soft andsometimes with loose stool at the end of the eighth week. Compared to the normalgroup. at the end of the eighth week, we can see the rats in model group are rich inadipose tissue, liver volume increasing, and we can have the feeling of greasy whentouch the section. When compare the model group to normal group at the end of theeighth week, we can see the increase of weight and wet liver weight of rats (P<0.05),increase of blood serum ALT、AST、TC、TG and liver TC、TG(P<0.05);The liverpathological histology test of the rats in model group shows that more than1/3of theliver cell has fatty degeneration, which indicates that the models of early fatty liver inrats have been made successfully.Ⅱ. Compared to the normal group, each index of rats’hemorheology in modelgroup changed evenly at the end of the eighth week, including blood viscosity, plasmaviscosity, aggregative index of red blood cell, fibrinogen, packed cell volume,platelet aggregation rate (P<0.05) and speed up in blood sedimentation(P<0.05),which indicates the abnormal of hemorheology in early fatty liver.Ⅲ. Compared to the model group, the rats in Quzhi Ruangan Decoction relievedin pathological changes of liver fatty degeneration, abnormal liver function and bloodlipids elevated to some extent at the end of the eighth week. which indicated that theQuzhi Ruangan Decoction has good intervening role to disease development of earlyfatty liver in rats. Ⅳ. The hemarheology index of the rats of Quzhi Ruangan Decoction changed invarious degrees at the end of the eighth week. Compared to the model group, theindex decreased in blood viscosity, aggregative index of red blood cell, fibrinogen,packed cell volume, platelet aggregation rate (P<0.05) and slow in bloodsedimentation(P<0.05), and the plasma viscosity decreased to some extent but nostatistic meaning. which indicated that the Quzhi Ruangan Decoction has goodintervening role to the abnormal hemarheology of early fatty liver in rats.Conclusion:Ⅰ. Improving the formula high fat forage (adding10%of lard oil and2%ofcholestenone to normal animal forages) to feed rats for the typical early fatty livermodels at the end of the eighth week.Ⅱ. The obstacles of each hemarheology index of early fatty liver in rats causedby high fat forage indicated.Ⅲ. Quzhi Ruangan Decoction can not only improve the changes ofliver dysfunction, lipid metabolic abnormalities of liver, but also improve eachhemarheology indexes abnormal of the rats’ model of early fatty liver cause by highfat forage effectively. which indicates that Quzhi Ruangan Decoction may cure thefatty liver by improving the index of hemorheology and the mechanism of it remainsfor further experimental research.Ⅳ. There is a relationship among fatty liver, blood stasis syndrome of traditionalChinese medicine and blood stasis rheology, but the mechanism of it remains forfurther experimental research.
