| Objective: Ulcerative colitis is a chronic and nonspecific inflammatory disease often involving the rectum and colon which is not yet elucidated in etiology and pathogenesis.In this study,we observed the general situation,the index of disease activity index(DAI),the histopathological changes,the content of inducible nitric oxide synthase(i NOS)and the colon CD40 expression levels in the tissue in the UC rat model,and explore the mechanism of Huazhuojieduyuyang decoction for the treatment of ulcerative colitis,and provide experimental basis for the clinical application of this prescription.Method: Sixty clean-grade healthy male Wistar rats(body weight200g-220g)were fed for 1 week adaptively and then randomly divided into blank control group(NG group),model group(M group),mesalazine group(MS group),high dose of traditional Chinese medicine group(C1 group),middle dose group(C2 group)and low dose group(C3 group),6 in each group.Except for the normal feeding in NG group,all rats in other groups were reproduced UC rat model by 2,4,6-trinitro-benzene-sulfonic acid(TNBS)/ethanol combined modeling.Macroscopic observation two pre made model rats after execution of colon organization and the building after the death of four rats colon tissue,are visible colonic mucosal hyperemia edema,with ulceration,determine the building success.NG group continued routine feeding,M group was given saline,MS group was given Western drug mesalazine suspension gavage,C1,C2 and C3 group were given high,medium and low concentrations of three kinds of traditional Chinese medicine gavage.The changes of general conditions such as food intake,body weight,hair color,mental status and stool performance were observed continuously for two weeks and DAI scores were performed.After the observation of the rats in each group,fasting could not help but water for 24 hours,lidocaine anesthesia,femoral artery blood,chemical determination of i NOS levels in the blood of rats,immunohistochemical detection of lesions in the colonic mucosa of CD40 The expression level.Results:1.General observation and DAI scoring results NG group rats are in good spirits,with bright and smooth fur,active and active,like foraging,eating as usual,gaining weight,and having normal urine.The rest of the rats in the model after the successful replication,there are varying degrees of stool frequency increased,with mucus pus and blood,apathetic,fur dull,messy,dislike activities,eating less,thin body.After treatment,the rats in group C1,C2,C3 and MS except M group had the most improvement,the most obvious improvement was in group C1,while there was no significant difference in group MS and group C2,which were better than C3 group.DAI score results show:C1 group,C2 group,C3 group and MS group were lower than M group,the difference was statistically significant(P<0.05);C1 group was lower than C2 group,C3 group and MS group,the difference was statistically significant(P<0.05);C2 group,MS group were lower than the C3 group,the difference was statistically significant(P<0.05);The difference between C2 group and MS group was not statistically significant(P>0.05).2.histopathological observation of colon NG group rats colon tissue mucosal surface is smooth,colour and lustre is normal,mucosal epithelial integrity,continuity,glandular structure clear,submucosal no inflammatory cell infiltration,muscle layer structure without exception;In the M group,mucosal full-thickness ulcers were observed,mucosal glands disappeared,and mucosal and submucosal tissues around the ulcers were hyperaemia and edema,submucosa was thickened,and a large number of inflammatory cells infiltrated;The colonic mucosa of group C1 was smooth and mild edema,the structure of gland was clear,a small amount of lymphocytes scattered in the lamina propria,and there was no abnormality in submucosa and muscular layer;In C2 group,colonic mucosa was less smooth,erosion or ulcer improved,the number of goblet cells was more,the infiltration of neutrophils and chronic inflammatory cells in intima decreased,and crypts decreased;C3 group showed intestinal mucosa less smooth,the surface epithelium is not continuous,crypt structure changes,a large number of visible intrinsic membranous lymphocytes and plasma cells infiltration;MS group showed intestinal mucosa less smooth,surface epithelial continuous,a small amount of lymphocytes visible in the intrinsic membrane,plasma cell infiltration,reducing the number of crypts,a small amount of visible structural changes,a large number of goblet cells.3.Indicator detection i NOS content: Compared with NG group,the contents of i NOS in serum of other groups increased,the difference was statistically significant(P<0.05);numerical value of C1 group was lower than the M group,C3 group,MS group and C2 group,the difference was statistically significant(P<0.05);numerical value of C2 and MS groups were lower than those of C3 and M groups,the difference was statistically significant(P<0.05);numerical value of C3 group was lower thanthat of M group,the difference was statistically significant(P<0.05);There was no significant difference between C2 group and MS group(P>0.05).CD40 expression level: Compared with NG group,the CD40 density of colonic mucosa in group C1,group C2,group C3,group MS and group M increased,the difference was statistically significant(P<0.05);The increase of CD40 density in colonic mucosa in group C1 was lower than that in group MS,C3,C2 and M(P<0.05);There was no significant difference in density of CD40 between MS group and C2 group in colonic mucosa(P>0.05);The CD40 density distortions in colonic mucosa in C2 group and MS group were lower than those in C3 group(P<0.05).Conclusion: Taking Huazhuojieduyuyang decoction can improve the general situation of UC rats.The comparison between the treatment groups,Chinese medicine high-dose group improved better than the other groups, suggesting that there is a certain relationship between the dose of Chinese medicine and the treatment effect.The mechanism may be related to the decrease of i NOS level in serum and the decrease of CD40 expression in colonic mucosa,so as to promote mucosal healing. |
