Objective:To investigate the expression of Bmi-1and MMP-2,E-cadherin proteins in transitional cellcarcinoma of bladder, andexplore therelationship and correlation studies of expression of themand the occurrence,development, invasion and metastasis of the bladder carcinoma.Methods:The46patholoqical tissueof the bladder transitional cellcarcinoma was demonstrated by surgerywith complete clinical data wereselected from the year of2006to2012. Adopting the expressions of Bmi-1and MMP-2, E-cadherin proteinson these pathological sections prepared byS-P immunohistochemical staining. The immunoassay results were analyzedbySPSS statistical software Program (version16.0), χ2testwas used to analyzethe relationship of expression of them and the occurrence, development,invasion and metastasis of the bladder carcinoma. The correlation analysis ofexpressions ofBmi-1and MMP-2, E-cadherinwascompletedbySpearman’srank correlation analysis, α=0.05was considered as satisticallysignificant value.Results:(1)The positive expression rate of Bmi-1proteinwas10.00%innormal bladder mucosa, and positive expression in bladder cancer at a rate of52.17%;Itspositive expression rate in Tis-T1phaseof TCCBwas37.04%,positive expression rate was73.68%in T2-T4phase;Positive expression rateofBmi-1protein was37.50%in TCCB at a low level, in the high-level TCCBpositive expression rate was68.18%; The positive expression rate of Bmi-1inlymph nodes metastasis group was81.25%, Without lymph node metastasisgroup rate was36.67%.The above differences of expression between thecomparison groups had statistical significant (P<0.05).(2)There is no expression in normal bladder tissue specimensa aboatMMP-2protein, positive expression in bladder cancer at a rate of73.91%; In Tis-T1phase of TCCB, Its positive expression rate was59.26%, the positiveexpression rate in T2-T4is94.74%.Positive expression rate ofMMP-2proteinwas58.33%in TCCB at a low level, in the high-level positive expression rateis90.91%;Group with lymph nodes metastasis positive expression of MMP-2protein rate was100%, the positive expression of group without lymph nodemetastasis rate was26.67%. The above differences of expression between thecomparison groups had statistical significant (P<0.05).(3)E-cadherin protein no abnormal expression in normal bladder mucosa,and the abnormal rate was45.65%in bladder cancer tissues; The abnormalrate of it In Tis-T1phase of TCCB was29.63%, abnormal rate of T2-T4phasewas73.68%; In the low-level TCCB,E-cadherin protein abnormal rate of20.83%, the abnormal rate was72.72%in the high-level TCCB; With lymphnodes metastasis group of abnormal rate was87.50%, the abnormal rate was23.33%without lymph node metastasis group. The above expressiondifferences between the comparison groups were statistically significant (P<0.05).(4)Bmi-1and MMP-2were positively correlated, the rs’value of0.621.Bmi-1And E-cadherin, MMP-2and E-cadherinr were negativelycorrelated, thers’value respectively were-0.492and-0.544, the aboveresulthad statistical significant (P<0.05).Conclusions:1. Bmi-1and MMP-2, E-cadherin positive expression inbladder transitional carcinoma is closely related to the pathological gradingand clinical staging. According to the experimental data show that when thebladder cancer differentiation reduced and the degree of invasion, metastasisincreased, and positive expression of MMP-2and Bmi-1rate also increasedsubsequently, this relationship was positively correlated, and E-cadherinpositive expression rate decreases, was a negative correlation.2.Bmi-1and MMP-2, E-cadherin intheprocessofdevelopment, invasionand metastasisofbladder transitionalcell carcinomamay havemutual coordinationrole.
