Objective To explore the relationship of MYH7,MYBPC3gene mutation and the ACE/DD genotype in LVH patients. From the level of molecular genetics looking for hypertensive LVH possible reasons, so as to provide certain theoretical basis for the prevention and control of hypertension LVH.Subjects and Methods A total of313cases were enrol1ed in the trial,213essential hypertension patients randomly selected from Heart ward in Jining Medical College affiliated hospital,100cases of normal Control selected from Health screening center. According to the2010the prevention and control of hypertension of guidelines in China, systolic blood pressure≥140mmHg or diastolic blood pressure≥90mmHg selected for hypertensive patients (1mmHg=0.133kPa); LVMI≥125g/m2in male or≥120g/m2in female standard in Left ventricular hypertrophy. According to the diagnostic standard score three groups, namely hypertension left ventricular hypertrophy group112cases, high blood pressure of left ventricular hypertrophy group101cases, normal Control group,100cases. Collect the peripheral blood specimen of research object is used to extract DNA and testing of the biochemical indicators. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) detection I/D polymorphism of ACE, Select ACE/DD genotype samples amplified MYH7, MYBPC3purpose gene and gene sequencing. Using SPSS13.0software to analyse the datas. There is significant difference when P value was below0.05.Results1. A total of313cases were enrolled in the trial, finally303patients had conducted a successful whole blood genomic DNA extraction and polymorphism analysis. Gene I/D site of ACE existed II、ID and DD three genotypes, The Control group with100cases, II、ID and DD genotypes were49%,39%and12%. EH-NLVH group with101cases, II、ID and DD genotypes were49.5%,36.6%and13.9%. EH-LVH group with102cases, II、ID and DD genotypes were20.6%,48%and31.4%.Control group'EH-NLVH group'EH-LVH group, D allele frequency distribution were31.5%,32.2%,55.4%. Control group and EH-LVH group, EH-NLVH group and EH-LVH group, genotype distribution have statistical significance(P<0.01). The distribution of D allele frequency of EH-LVH group were significantly increased than Control group and EH-NLVH group. ACE/DD genotype samples amplified MYH7, MYBPC3purpose gene has nogene mutation.2. Multiple linear regression analysi, LV, IVS, BSA, LVPWT and heart rate has a great influence on Left ventricular hypertrophy. The binary Logistic regression analysis, ACE gene DD genotype increased the risk of left ventricular hypertrophy (P=0.03), and was1.89fold than genotype II.Conclusion1. The I/D polymorphisms of ACE existed in the southwest of Shandong Province. The genotype and allele distribution accord with Hardy-Weinberg Equilibrium. ACE gene polymorphism have significant relationship with high blood pressure LVH, DD genotype vulnerable to left ventricular hypertrophy.2. MYH7,MYBPC3genes mutations and LVH hypertension has no obvious relation,DD genotype of ACE gene and MYH7,MYBPC3genes has no joint action.
