Objective To investigate the correlation of expression of eIF4E and PDGF in endometrial cancer through the method of immunohistochemistry. Explore the relationship and related characteristics of eIF4E and PDGF in endometrial cancer with between the clinical parameters.and provide the theories basis for the earlier diagnosis and treatment of endometrial cancer.Materials and methods Endometrial cancer specimens of paraffin embedding were collected from51patients and Specimens were confirmed by surgery and pathology,35cases of the pathological slices of paracancerous tissues of2cm organization were selected as control group.42cases of normal endometrium samples were obtained from uterus which were removed because of cervical cancer in situ or hysteromyoma as a control group.Clinical and pathological data were integrity. Preoperative radiotherapy and chemotherapy were not applied. The expressions of eIF4E and PDGF in endometrial carcinoma were detected by immunohistochemical followed by observation under light microscopy and statistical analysis of the data.Results1. The eIF4E protein located in the cytoplasm of endometrial glandular epithelium showed claybank. Out of51cases,46cases of eIF4E expression, the positive expression rate was90.2%while the positive expression rate in the peritumoral tissue and normal endometrial tissue were28.6%and4.8%respectively. The difference was statistically significant (P<0.05).2. eIF4E protein had no significant difference between differentiated tissue and undifferentiated tissue of endometrial cancer.3.The positive expression rate of eIF4E protein in endometrial cancer without lymphatic metastasis and with lymphatic metastasis group were78.9%and96.9%, the difference was statistically significant. 4.PDGF positive staining of the cytoplasm or nucleus showed claybank. The rate of positive expression in the peritumoral tissue and normal endometrial tissue were37.1%and23.8%, which is lower than the endometrial tissue. In51cases.36cases of PDGF protein expression, the positive expression rate was70.6%while the well moderately differentiated tissues and poorly undifferentiated tissues, the positive expression rate was93.8%, and60.0%. The difference was statistically significant.5.The positive expression rate of PDGF protein with infiltration of muscle layer group and without muscle layer group was higher than non-infiltration muscle layer group.6. The positive expression rate of PDGF protein with lymphatic metastasis group was higher than without lymphatic metastasis group.7.PDGF and eIF4E protein, the positive expression rate in endometrial carcinoma was significantly positively related (P<0.05).Conclusion1.The expression of PDGF and eIF4E increased, indicating that the two may be involved in the occurrence of endometrial cancer.2.When the tissue differentiation in endometrial cancer is lower and infiltration depth of muscular layer associated with lymph node metastasis, PDGF expression increased significantly, suggesting that PDGF promote invasion and metastasis of endometrial cancer.3.The infiltration depth to muscular layer associated with lymph node metastasis, eIF4E expression increased significantly, which eIF4E promote invasion and metastasis of endometrial cancer.4.PDGF may promote endometrial carcinoma cell expression and eIF4E may induce the expression of PDGF. Both of them are associated with the development of endometrial cancer.
