AIM:To investigate midkine(MK)level in serum of patients with esophageal squamous cell carcinoma before and after operation,detect the expression of MK protein in tissues and analyze its relationship with clinical stage, pathological features and biological behavior. Provide an objective basis for the diagnosis of esophageal squamous cell carcinoma, efficacy monitoring and assessment of prognosis.METHODS:Fasting venous blood of55patients with esophageal squamous cell carcinoma in chest Section II of Shanxi Tumor Hospital and30cases of healthy controls should be gathered from July2010to July2011.Automatic ELISA analyzer was used to detect the level of serum MK,while immunohistochemical method was used to detect the tumor tissue and corresponding adjacent normal tissue of MK protein expression.RESULTS:1.The average optical density(O.D.)of MK in serum of esophageal squamous cell carcinoma patients before operation was0.1006±0.0624,0.0455±0.0155in normal control group,0.0752±0.0267in postoperative control group.The MK levels in serum of esophageal squamous cell carcinoma patients before operation were significantly higher than normal and postoperative control group, statistically significant difference (P<0.001).2.The preoperative serum MK expression level of esophageal squamous cell carcinoma patients was no significant difference with age, gender, tumor size, TNM staging and lymph node metastasis (P>0.05),but significantly statistic difference with cell differentiation, depth of invasion, vascular involved (P<0.05).3.MK positive expression rate was58.2%(32/55) in tumor tissues, no expression in adjacent normal tissues.There was significantly statistic difference between two groups (P<0.001).4.MK protein expression in tumor tissue was no significant difference with age, gender, tumor size, TNM staging and lymph node metastasis (P>0.05),but significantly statistic difference with cell differentiation, depth of invasion, vascular involved (P<0.05).CONCLUSION:1.MK was overexpression in the serum and tumor tissue of esophageal squamous cell carcinoma patients, and related with the occurrence of esophageal squamous cell carcinoma2.Preoperative serum MK expression was significantly higher than normal and postoperative control group, which indicated that the high level of preoperative serum MK was caused by the tumor. MK may be used as the postoperative follow-up monitoring of tumor recurrence and metastasis of indicators.3.MK expression level in the serum and tumor tissue correlated with age, gender, tumor size, TNM stage and lymph node metastasis (P>0.05),meanwhile there were significantly statistic difference with cell differentiation, depth of invasion, vascular involving related(P<0.05).These showed that MK may promote tumor invasion and angiogenesis.
