Objective: To observe the EA pretreatment of different point group for liverdepression and spleen rat spleen cell apoptosis，the expression of BCL-2andBax proteins.To reveal the mechanism of EA pretreatment on the prevention ofliver stagnation and spleen deficiency.Methods:50Wistar rats were only male and female half were randomly dividedinto five gro ups according to body weight， male and female in each group offive male and female separ atee feeding.Grouped as follows:blank control group(A)，model control group(B)，“Taich ong+Sanyinjiao” group (C)，“Taichong+Sanyinjiao the Tianshu” group (D)，“Taichong+Sanyinjiao+on the giantvirtual”group(E).7days before the experiment，electroacupuncture prec onditioning in rats in group C and D，E，20min，while ripping the A and B rats.8to22days，rats with chronic stress (unpredictable) modeling，the method ofselecting bound，off food，water， black and white reversal，45-degree warmwater swimming, random number table， arrange the order of addition to the blankgroup. Each method implementation，imple mentation，modeling，a total of15days.Spleen cells Bcl-2and Bax protein expression observ ed by immunohistochemicalmethods,as well as to observe the group of spleen apoptosis rate by TUNEL assay.Results:1.Normal control rats the appearance of behavior no obvious abnormalities. Incontrast， mod eling the7th day,the normal build module rats that showed thelevel of activity weakened，eati ng less，easy to wake up, stool softened；ac upuncture rats pretreated with no significant cha nge. Modeling13d model grouprats showed normal get together and even depression， apath y，the battle toreduce food intake， stool into thin pond， increased frequency of defecation，the hair becomes dry the roots yellow， diminished activity， tired of lying； acupuncture rats pretreated with no significant change.2.With the blank control group, model control group spleen Bax protein expressionof Bcl-2protein downregulation, the change was statistically significant (P<0.05)；pretreatment with the acupuncture of different acupoints group，the modelcompared to the control group，spleen cells Bax protein expression increasedand Bcl-2protein downregulation，the change was statistically significant (P<0.05)；the acupuncture pretreatment acupoints group with the control group，the spleen cells of Bax protein and Bcl2protein expression fluctuations ofno statistical significance.3.With the blank control group，model control group，spleen cells from apoptosisrate increased，the change was statistically significant (P <0.05)；pretreatmentwith the acupuncture of different acupoints group，model control group spleencell apoptosis rate increase，the change was statistically significant (P <0.05)；compared to the pretreatment of the various acupuncture acupoints group withthe control group，the apoptosis of spleen cells was similar in no statisticallysignificant.Conclusion:1.EA pretreatment liver stagnation and spleen deficiency syndrome have asignificant preventive effect.2.EA pretreatment on the prevention of apoptosis of spleen cells wassignificantly inhibited.3.Stagnation and spleen deficiency syndrome can lead to apoptosis of spleen cells.
