Differential Expression Of Th Cytokine Between Chronic Hepatitis B With Liver Stagnation And Spleen Deficiency And Spleen And Stomach Damp-Heat And Its Correlation With H2BE Gene

Objective To analyze the cellular immunological difference between CHB with liver stagnation and spleen deficiency syndrome and spleen and stomach damp-heat syndrome based on the Th cytokines IL-2,IL-4,IL-10 and INF-?.On this basis,make clinical verification of H2BE that screened by high throughput in the early stage.Finally,make a preliminary study on the correlation of Th cytokines and H2BE expression between the two syndromes.Methods A total of 22 patients with CHB are enrolled in accordance with the syndrome of liver stagnation and spleen deficiency and spleen and stomach damp-heat,and 9 healthy volunteers are included at the same time.Extract the peripheral blood separately of them as experimental specimen.Detect the levels of IL-2,IL-4,IL-10 and INF-?by ELISA,and detect the level of H2BE by RT-qPCR,SPSS24.0 is used for data analysis.GeneMANIA database is used to construct protein interaction network correlation map.Results Compared with the healthy group,the expression levels of IL-2,IL-4 and INF-?are increased in both syndromes of CHB(P<0.05),the level of IL-10 in liver stagnation and spleen deficiency syndrome is higher than that in healthy group(P < 0.05),the levels of IL-4,IL-10 and INF-?have significant difference between the two syndromes(P < 0.01),there is no difference of IL-2 expression between the two syndromes(P>0.05).Compared with the healthy group,the level of H2BE is increased in both syndromes of CHB(P < 0.05),and there is a significant difference between the two syndromes(P < 0.01).H2BE,IL-2,IL-4,IL-10 and their relevant molecules are related through physical interaction,co-expression,co-localization and pathway.Conclusions There is a discrepancy in Th cellular immune function between CHB with liver stagnation and spleen deficiency syndrome and spleen and stomach damp-heat syndrome,cellular immune suppression exists in both syndromes,notably in liver stagnation and spleen deficiency syndrome,this may be related to the formation of immune tolerance in CHB with liver stagnation and spleen deficiency syndrome;IL-4 and IL-10 may be the modern immunological gist for the difference between the two syndromes.H2BE gene may be the biological gist for the difference between liver stagnation and spleen deficiency syndrome and spleen and stomach damp-heat syndrome of CHB.The variation of H2BE gene level may be related to the activation of CD4 + T cells.H2BE gene is associated with IL-4 and IL-10,in which,GATA3 may be the intermediate to participate in the related pathway,this result provides a potential research target for subsequent study of signal pathway.
