ObjectiveTo investigate the effects of Duonang Decoction on the serum ho rmone levels, morphology of ovary and endometium as well as the end ometrial receptivity of infertility rats caused by polycystic ovary syndrome (PCOS) with hyperandrogenism (HA), and also to discuss it s therapeutic effect as well as the possible mechanism. And it will provide a new clinical rationale for treatment of polycystic ovari an syndrome.MethodsTo develop a model of PCOS in9-day-old female rats were divide d into model group(n=160) and normal group(n=40) randomly. The rats in model group were injected with testosterone propionate on the b ack of the neck for1.25mg per rat as Dr.Yu did. Then the model gr oup rats were divided into four groups:control group (0.9%normal saline), positive group (clomifene citrate100mg/kg/d), high dosage decoction group (Duonang Decoction68.66g/kg/d), low dosage decoct ion group (Duonang Decoction34.33g/kg/d), and each group had40ra ts. For the positive group,2days for a estrous cycle were fed by drugs, the rest days were fed by0.9%normal saline. Other groups w ere fed as ruled for10days. Tested the serum hormone levels (testos terone, follicle-stimulating hormone) of the abdominal aorta blood collected from15rats of each group by enzyme-linked immunosorbent assay, and observed morphology changes of left ovaries under the m icroscope. Then the rest were mated with male rats (4:1)and were ch ecked the pregnant everyday. Took first day of pregnant as D1and t hen killed the rats at D5. Dividing the left part of the uterines i nto two parts, one parts were observed morphology changes under the microscope, for the other scanning electron microscopy (SEM) were used to explore the development of the pinopodes.Results1Compared with control group, the testosterone(T) levels of the hi gh dosage decoction group, normal group and low dosage decoction gro up and positive group all reduced with statistical significances (p<0.05), especially the high dosage decoction group (p<0.01). About f ollicle-stimulating hormone (FSH), the levels of it were all increa sed in treatment groups compared with control group, especially the high dosage decoction group (p<0.05) which was nearly the same as n ormal group.2Compared with normal group, the quantity of follicles of the othe r four groups all decreased. Among the four groups, the results of control group, positive group and high dosage decoction group had s tatistically significant (p<0.05), while no statistical significan ce was found in low dosage decoction (p>0.05). When compared with the control group, the follicle diameter and the thickness of ovaria n theca cell layer were with no difference in all the groups (p>0.05); the thickness of ovarian granule cell layer in normal group and high dosage group were thicker (p<0.01)。When observed under the m icroscope, typical structure, thick ovarian granulose cell layer, a bundant corpus luteum and follicles can be found in slices of the o varies of the normal group, the low dosage decoction group, the hig h dosage decoction group as well as the positive group. However in control group, atypical structure, thin ovarian granulose cell laye r, less corpus luteum and follicles were always seen.3Under the microscope, the thickness of endometrium in treated gro ups were thicker when compared with the control group (p<0.05). The well-developed secretory phase endometrium can be found in normal g roup, high dosage decoction group, low dosage decoction group and p ositive group. While only poor-developed proliferating phase endome ntrium can be found in control group.4When observing under SEM, endomentria of normal group, high dosag e decoction group, low dosage decoction group and positive group ha d mature pinocytosis, while no mature pinocytosis on the endomentri a of control group.ConclusionDuonang Decoction can decrease the T levels and increase the FS H levels and the quantity and the quality of follicles of the infer tility rats caused by PCOS with hyperandrogenism. So it can adjust and balance the function of hypothalamic-pituitary-ovary axial syst em. In addition, Duonang Decoction can also improve endometrial rec eptivity of PCOS rats.
