Vancomycin is a glycopeptide antibiotic with a complex structure. It is produced by a kind of streptomyces, which can specifically inhibit peptidoglycan biosynthesis. Vancomycin can be used when other antibiotics against bacteria is invalid. Vancomycin is mainly used for system infection and intestinal infection that caused by staphylococcus aureus (including penicillin resistant strains and new nystatin resistance Strains). So known as the last line of drugs.In this thesis, the vancomycin filtrate after microfiltration and nanofiltration is used as raw material. The purification conditions of vancomycin are investigated to develop a process with high recovery and better purification effect. During the present experiment, anion resin Amberlite FPA53is used to remove pigment and regulate the pH of the raw material. Then, six resins are selected and test the adsorption isotherm, the effects of pH, concentration, flow rate and elution conditions on adsorption are studied. As a result, the best adsorption resin (Amberlite XAD1600N) and appropriate technological conditions are obtained.For further separation and purification of vancomycin crude product, resin Homo30RPC-300is used. The effect of temperature on separation is investigated. After optimization, vancomycin with purity more than96%(detected by chromatography) is obtained. Through this research, a more suitable industrial process than present industrialized process is developed.The new process can be used to obtain vancomycin product with high purity and stable quality.
