Alkaloids, which provides many pharmacologically active compounds, belongs to one kind of plant secondary products. Datura Stramonium L. belongs to Datura, which is Mandalas of annual herb plants. The whole plant can product Hyoscyamine and Scopolamine, which is one of the resource plants of Tropane alkaloids. Scopolamine is basal important medicine, and widely used in clinic. There is a large demand of scopolamine in market.The stem of Datura stramonium L. was used as explants, and inducted its callus to compare the effects on callus induction and generation among the different concentration of phytohormone and to extract alkaloids from a callus cultivating.It settles a firmly foundation for metabolic engineering via leaf of molecular breeding of Tropane alkaloids to be realized using plant secondary metabolic engineering.Compares the different concentrations of hormone and other external conditions brugmansia callus induction influence, The results showed that the ideal medium for induction of callus were MS+2,4-D1.0mg.L-1; the ideal medium for generation of callus were MS+2,4-D0.5mg.L-1;The study of the different light and temperature conditions on the growth of callus shows that constant temperature of24℃, light for the2000-30001x,12h·24h-1make the callus grow so good. By comparing the influence of the different carbon sources on the growth of callus, the results showed that sucrose is an ideal carbon source. The generation of its callus study finds, the liquid culture of Datura callus growth rate of30d is30.13%, and the solid culture of Datura callus growth rate of438%. Qualitative and quantitative analysis was done to hyoscyamine and scopolamine which were extracted from Datura stramonium L. By high performance liquid chromatography (HPLC) detection datura callus tissue of alkaloid content, the results show that by callus extraction of alkaloids contains drug scopolamine and the drug scopolamine. This results strongly proves, can quickly by tissue culture technology to get a lot of Datura callus containing Tropane alkaloids, which has a very important significance and commercial value.
