| The Japanese Hokkaidao special product small tree(lonicera caerulea L.Var.emp-hyllocaryx NAKAI)belongs the plant of Caprifoliaceae.It is a very valuable wild fruit germplasm resources with high nutritional, medicinal and economic values. This test iexplored the conditions including different materials, substrates and hormones and related factors of cuttings rooting and transplanting which was on the basis of the techniques of fast multiplication for the tissue culture of HASKAOP.The main results of this research were as follows:1. The cutting rooting research(1) The best cutting material for the cutting rooting is the half lignification stems of central section, the rooting rate was82.67%and the average root length was3.06cm.(2) The orthogonal experiment showed that two combinations were better for the cutting rooting. The stems were soaked with200mg/L IBA for10minutes and then planted in base soil with vermiculite:peat soil=2:1or soaked with100mg/LIBA for60minutes and then planted in the base soil with vermiculite:peat=2:1.(3) The stems after treated by200mg/L IBA got the rooting rate of88.67%under the second treatments on cutting of HASKAOP.(4) Both in the spring and autumn could obtained high rooting rate for the cutting rooting.The rooting rate were89.33%and84.00%.2. The transplanting research(1) The the survival rate of HASKAOP for transplanting was75.33%at the depth of2cm and compacted the substrate.(2) The HASKAOP got the maximum survival rate under the method which was adopted water domestication method for hardening plantlets before transplanting.The survival rate was65.0%and new leaves number was2.31under this way.(3) The survival rate was59.0%under the substrate of vermiculite:peat=1:1and the number of new leaves was up to3.34.It is the most suitable substrate for transplanting.(4)100mg/L ABT could been used for HASKAOP before transplanting to deal with the tissue culture.But the transplanting efficiency of HASKAOP had barely improved with using any hormones.(5) The optimal time for transplanting of HASKAOP was spring and the survival percent was62.0%. |
