Aims to solve the problem of long rooting time,irregular rooting and low root ing rate in the asexual rapid propagation of Sabina chinensis species.Using Sabina chinensis 'Lanta',Sabina chinensis 'Diecui',Sabina chinensis 'Fenggui',Sabina chi nensis 'Rongtuan',Sabina chinensis 'inhua'as test materials.four kinds of growth re gulators(ABT-1,IBA,NAA,IBA:NAA=2:1)and each growth regulator with five concentrations(50,100,200,500,1000 mg·L-1)were studied on rooting effect,usi ng a completely randomized block design.The effects of cutting media and differen t tree ages on Rooting of 'Lanta' also be studied.On this basis,the anatomical and physiological mechanisms of adventitious roots in cutting process of Sabina chinen sis were studied in depth.Below are key research findings.1.Research on cutting propagation techniques(1)The highest values of membership functions of rooting indexes of the Sa bina chinensis 'Lanta',Sabina chinensis 'Diecui',Sabina chinensis 'Fenggui',Sabin a chinensis 'Rongtuan' and Sabina chinensis 'Jinhua' were 1000 mg·L-1 IBA:NAA=2:1 soaked for 10s(0.678),200 mg·L-1 IBA:NA=2:1 soaked for 4h(0.685),1000 mg·L-1 IBA:NAA=2:1 soaked for 10s(0.497)and 100 mg·L-1 soaked for 4h(0.378)and 500 mg·L-1 IBA:NAA=2:1 soaked for 4h(0.494).(2)The annual cuttings of Sabina chinensis were mainly rooted in callus,foll owed by cortex and their mixture was the least.(3)The genetic characteristics of each variety are different,of which Sabina chinensis 'Diecui' has the highest rooting rate,Sabina chinensis 'Rongtuan' has the lowest rooting rate.(4)For the cutting propagation of Sabina chinensis 'Lanta',among rooting me diums,perlite>river sand>river sand:grass charcoal=1:1>river sand:grass charcoal:perlite=1:1:1,and there was no significant difference in the rooting effects of the st ock plants age between 10 and 35 years old.2.Anatomical studies on rooting process(1)Sabina chinensis rooted slowly and unevenly.(2)No thick-walled ring cells obstructing rooting were found in the stem sect ions of the five Sabina varieties.(3)Sabina chinensis 'Lanta' has multiple sites to form the latent root primord ia,however,its rooting method is still dominated by induced root primordia which also formed at multiple sites.3.Studies on physiological indicators(1)The effect of soluble sugar and starch on the rooting of Sabina chinensis cutting propagation was not significant,but the soluble protein had significant effe cts.(2)According to the change of soluble protein content,the rooting of Sabina c an be roughly divided into callus induction period(0?20 days),callus formation p eriod(20-40 days),adventitious root induction period(40-60 days),adventitious r oot formation period.(60-80 days),adventitious root elongation(after 80 days).(3)The change of PPO activity in the rooting process was not obvious,but POD activity changed obviously and mainly played in the early rooting stage that increased rapidly from 0 to 20 days and decreased sharply in 20 to 40 days.(4)The effects of IAA,ZR,IAA/ABA and IAA/ZR on the rooting of 'Diecu i' cuttings were not significant,the effects of ABA,GA3 and IAA/GA3 were signifi cant and mainly affected 0-40 days after cutting.When IAA content is higher tha n ABA content and lower than ZR content and GA3 content,the more conducive t o the formation of adventitious roots.(5)POD activity and endogenous hormones mainly regulate the formation of adventitious roots rather than growth.Rooting mechanism of Sabina chinensis cutting was studied from the perspectiv es of anatomy and physiology.By analyzing the dynamic changes of cuttings'morph ology,transverse section and physiological indexes during rooting.
