Study On Anesthesic Quality,Pharmacoki

There is no ideal intravenous anaesthetics specially for minipigs now. Ketamine, xylazine and midazolam are widely used for premedication, chemical restraint, and the induction and maintenance of anesthesia in swine. In the present study, according to the physiology characteris of minipigs, we combine the three anesthetics into a compound prescription for minipigs-Shumianning Ⅱ.We evaluate the effects of Shumianning Ⅱ administered by bolus intravenous injection on quality of clinical status of anesthesia, hemorheology, respiratory function and functions of liver and kidney in miniature swine,and study the pharmacokinetics of Shumianning.According to the first experinment, induntion anesthesia with Shumianning Ⅱ was quick and smooth, the stage of surgical anesthesia was long and smooth, recovery was quick and smooth. During the period of anaesthesia, effects of immobilization and analgesia is good; HR, NABP, RR, RT and PETCO2were all in the normal physiologic range. Only SpO2at1min and5min were were lower than the normal physiologic range. All the biochemical parameters were in the normal physiologic range. We can concluded that the effecs of Shumianning Ⅱ on the cardiovascular and blood chemistry was small, temporal respiratory depression was appeared.According to the study on the pharmacokinetics of Shumianning Ⅱ in minipigs, we knew that ketamine, xylazine and midazolam were best described of nonassimilation two-compartment open model and the typical pharmacokinetics parameters of the three drugs were as follows:Ketamine Vss=3.177±0.720L·kg-1, T1/2a=3.685±0.462min, T1/2β=77.882±12.699min, AUC=155.792±11.658μg·min·ml-1, CL=0.115±0.025L·min-1·kg-1; Xylazine Vss=4.046±0.807L·kg-1, T1/2a=4.927±1.640min, T1/2β=88.083±37.860min, AUC=74.116±25.380μg·min·ml-1, CL=0.0388±0.0181L·min-1·kg-1; Midazolam Vss=1.175±0.242L·kg-1, T1/2a=3.359±1.741min, T1/2β=42.489±13.088min, AUC=10.951±2.190μg·min·ml-1,CL=0.022±0.004L·min-1·kg-1.Effects of Shumianning Ⅱ administered by intravenous injection on cardiovascular in cystotomy in Bam a minipigs is small, temporal respiratory depression was appeared. Hypothermia was appeared after90min.These minipigs were well sedated. Minipigs had non pain reaction on every operative procedure. Sarcous tension hasn’t been perceived in every operative procedure.Therefore, Shumianning Ⅱ meets clinical operation requirement.
