Studies On Screening Dorminant Cultivars’ After-ripening Days And Casing Overlay For Pleurotus Eryngii

Under the different conditions,22strains were compared by the mycelium cultured and cultivated experiments which were from different regions. And these different conditions were temperature, pH, cultivation materials of various nitrogen content. The biological characteristics were analyzed and compared, such as mycelium growth characteristics,the time of primordium formation,the time of harvesting,cultivated period and the characteristics of fruiting body. Other traits were also compared which were yields and biological efficiencies of different cultivars. The best strain was evaluated by synthetically analyzed. The different after-ripening days of the best strain were compared and the optimal after-ripening day was selected. And then these different cultivated bags were transfered to the cultivated rooms. The color,pH and the activity of ligninase were obsurved and measured which were belonged to the raw materials in the cultivated bags. Comparing the cultivative characteristics,biological efficiency and so on,then,the optimal after-ripening day was selected. The yield of Pleurotus eryngii which has finished one flush was improved by the use of cultivation method that was casing overlay and the twice management for the cultivative bags was achieved. Using the single factor analysis and orthogonal tests,the thickness,proportion and humidity of the covering materials were studied for the optimum conditions preliminarily. In order to offer the better strain,optimal after-ripening day,explored a new methods to manage the after-harvesting cultivated bags,the experiments were did,and these could offer theory basis for production. The results showed that:1. The screening of the dominant Pleurotus eryngii strainIn order to screen out the optimal Pleurotus eryngii strain for industrial cultivation. The biological charactristics of22strains of pleurotus eryngii were studied and compared under different temperature,pH and raw materials of different nitrogen content,which included mycelial growth potential,antimicrobial activity, time of primordium formation,harvesting time,crop cycle and biological efficiency. The results showed that:the strain of Xing1222was dominant strain and more adaptive for production practice,whether the hypha growth,the ability for antimicrobial,or the yield.And its optimium conditions were like these:the temperature was23℃,and pH was6.5,the nitrogen content of cultivation meterials was0.98%. And the highest biology efficiency was96.6%.2.The optimal after-ripening days of the dominant Pleurotus eryngii strainTake Xingl222strain as the tester strain which was got by screening experiments,the optimal after-ripening days were studied.After the hypha sprawling the cultivation bags,set the after-ripening days respectively which were0d,1d,2d,3d,4d,5d,6d,7d,8d,9d,10d,12d,15d,20d,25d. Compared with the time of primordium formation, harvesting time,crop cycle,the length of the stipe,the density of the fruiting body comprehensively,it was found that the optimal after-ripening days of Xing1222was8days,the highest biological efficiency was92.5%. And at that time,the materials’pH of cultivation bags was6.5,the color was oyster white,the laccase activity was33.6U/mL and the manganese peroxidase activity was27.1U/mL.3. The application of casing overlay cultivationThe optimal thickness for the covering material was3cm,the biological efficiency was61.9%,it’s higher than other thicknesses;when the proportion was3:1(peat moss/calcium carbonate),the biological efficiency was62%;and when the optimal humidity was85%,the biological efficiency of the second flush was62.7%,it’s the highest among the tests. Then the orthogonal tests were did,after variance analysis,combining all the datas,it was confirmed that A3B5C4was optimal combination.In this combination,the thickness of coverig materials was3cm,the ratio of covering materials was1:4(peat moss/calcium carbonate),the humidity of materials was85%. Under this condition,the time of primordium formation was7d,harvesting time was8d,the diameter of pileus was8.7,the length of the stipe was13.9cm,the yield was419.7g,and the biological efficiency was64.7%.
