This experiment was conducted to determine the effects of the stress by thecombination of molybdenum and cadmium on red blood cell injury and spleen-relatedgene expression in goat.63healthy goats were choosed and randomly divided to7treatments.Treatments consisted of control group(0+0), low cadmium lowmolybdenum group(0.5mgCd kg-1BW+15mgMo kg-1BW), low cadmium middlemolybdenum group(0.5mgCd kg-1BW+30mgMo kg-1BW), low cadmium highmolybdenum group(0.5mgCd kg-1BW+45mgMo kg-1BW), high cadmium lowmolybdenum group(1mgCd kg-1BW+15mgMo kg-1BW), high cadmium middlemolybdenum group(1mgCd kg-1BW+30mgMo kg-1BW), high cadmium highmolybdenum group(1mgCd kg-1 BW+45mgMo kg-1BW), each group had threerepetition. CdCl2and [(NH4)6Mo7O24·4H2O] as source of cadmium and source ofmolybdenum, respectively. The goats from control group were drenthedcorresponding quantitative deionized water. Blood samples were collected on0th,10th,20th,30th,40th and50th day in experimental period from every group afterovernight fasting to extract red cell membrane, then relevant indexes were determined.Spleen tissue were collected from3goats from each group after they were slaughteredon0th,25th,50th day to determine expression quantity of apoptosis gene Bcl-2, Bax,Cyt c, Caspase-3, Smac and gene of copper protein CP, in addition, changes inhistopathology and ultrastructure of spleen were detected by light microscope andtransmission electron-microscope, respectively.①Compared with the control group,the levels of MDA、XOD、LDH in red cell membrane from each group weresignificantly higher while ATPase、GSH-Px、GST、ALP were lower on50thday(P<0.01), T-AOC had no statistical significance (P>0.05);②Compared with thecontrol group, the expression quantity of Bcl-2, Cyt c, CP mRNA in spleen from eachgroup was significantly lower (P<0.05), while Caspase-3mRNA from each groupwas significantly lower except the expression in low cadmium low molybdenumgroup (P<0.05), Bax mRNA from low cadmium in molybdenum and high cadmiumhigh molybdenum group was higher (P>0.05), Smac mRNA had no statisticalsignificance on50th day;③Compared with control group, the volume of spleniccorpuscle decreased, hemosiderin deposited; the number of lymphocytes decreasedsignificantly, apoptosis body, vacuolation of cytoplasm and organelles from the spleentissue of high cadmium high molybdenum was visible on50th day;The results showed that the activities of antioxidant enzymes in red cellmembrane decreased and oxidant enzyme increased under stress from thecombination of molybdenum and cadmium, molybdenum and cadmium could involvethe injury of spleen through apoptosis manner which was mediated by mitochondrialpathway, and molybdenum and cadmium represented synergistic effect.
