This experiment was conducted to determine the effect of stress from the combinationof molybdenum and cadmium on free radicals mitochondria and mRNA expression in liverfrom goats.63healthy goats were divided into the following seven groups randomly:control group (which was drenched equal amount of deionized water), low-dose cadmiumcombined with low-dose molybdenum group (15mg Mo·kg-1·BW,0.5mg Cd·kg-1·BW),low-dose cadmium combined with moderate-dose molybdenum group (30mgMo·kg-1·BW,0.5mg Cd·kg-1·BW), low-dose cadmium combined with high-dosemolybdenum group (45mg Mo·kg-1·BW,0.5mg Cd·kg-1·BW), high-dose cadmiumcombined with low-dose molybdenum group (15mg Mo·kg-1·BW,1mg Cd·kg-1·BW),high-dose cadmium combined with moderate-dose molybdenum group (30mgMo·kg-1·BW,1mg Cd·kg-1·BW), high-dose cadmium combined with high-dosemolybdenum group (45mg Mo·kg-1·BW,1mg Cd·kg-1·BW). The experimental period was50days, blood collection were carried out on0day,10days,20days,30days,40days and50days; samples were obtained on0days,25days and50days by killing3goats fromeach group respectively. Subsequently, further experiments were conducted to observe theeffect of different Mo-Cd concentration on blood lipid, liver function, liver mitochondriafunction and mRNA expression in hepatic gene in goats. The results are as follows:1. Compared with control group, serum ALB, CP, Ch and TG in experimental groupsare significantly decreased(P<0.05); As time went on, serum AST increased significantly(P<0.05), moreover, tendency in groups with high Mo-Cd concentration are obvious thanthose with low Mo-Cd concentration.2. In experimental groups, as time went on, liver mitochondria T-AOC and SOD aresignificantly decreased(P<0.05), MDA, NO and NOS are significantly increased(P<0.05);Moreover, with the increase of concentrations of Mo-Cd, tendency wereincreasingly obvious.3. Compared with control group, liver Caspase-3, Smac and mRNA were increasedsignificantly(P<0.05), moreover tendency in groups with high Mo-Cd concentration areobvious than those with low Mo-Cd concentration.4.Through the observation of hepatic pathological slices in Mo-Cd group of theexperimental groups,the cells of liver showed swell and intercellular space become blurwith haemorrhage,vacuoles with different sizes within the cytoplasm, the severity of injuryin every experimental group is accelerated as the increase of time and dose ofMo-Cd;Observe the ultrastructure of high-dose cadmium combined with high-dosemolybdenum group at the50days:degeneration and necrosis of nucleus, mitochondriaswelling, reducing and vacuolation.Results showed that Mo-Cd have toxic effect on goats in liver apparently,combination of Mo and Cd disturbed metabolism of free radicals mitochondria in liver andincreasing of mRNA expression in hepatic gene in goats.
