This experiment was conducted to determine the effect of stress from the combinationof molybdenum and cadmium on serum free radical and expression of liver apoptosisralated gene in goat, which was in order to explore the toxicology relationship ofmolybdenosis and cadmium poisoning. This experiment was choosed the ammoniummolybdate [(NH46Mo7O24·4H2O] as a source of molybdenum and cadmium chloride(CdCl2) as a source of cadmium in this experience.63healthy goats were divided into thefollowing seven groups randomly: control group (which was drenched equal amount ofdeionized water), low-dose cadmium combined with low-dose molybdenum group (15mgMo·kg-1·BW,0.5mg Cd·kg-1·BW), low-dose cadmium combined with moderate-dosemolybdenum group (30mg Mo·kg-1·BW,0.5mg Cd·kg-1·BW), low-dose cadmium combinedwith high-dose molybdenum group (45mg Mo·kg-1·BW,0.5mg Cd·kg-1·BW), high-dosecadmium combined with low-dose molybdenum group (15mg Mo·kg-1·BW,1mgCd·kg-1·BW), high-dose cadmium combined with moderate-dose molybdenum group(30mg Mo·kg-1·BW,1mg Cd·kg-1·BW), high-dose cadmium combined with low-dosemolybdenum (45mg Mo·kg-1·BW,1mg Cd·kg-1·BW). The experimental period was50days,blood collection were carried out on0day,10days,20days,30days,40days and50days;samples were obtained on0days,25days and50days by killing3goats from each grouprespectively. Subsequently, further experiments were conducted to observe the effect ofstress from combination of different doses molybdenum and cadmium on serum freeradical metabolism, ruminal environment and expression of liver apoptosis ralated gene ingoat. The results are as follows:1. As time went on, the activity or content of NO, NOS and MDA in the poisonedgroups’ serum were increased significantly when compared with the control group (P<0.05or P<0.01), while the corresponding indexs of T-AOC and T-SOD were decreasedsignificantly (P<0.05or P<0.01). Moreover, with the increase of concentrations ofmolybdenum and cadmium, changing trends were increasingly obvious.2. As time went on, the pH value of rumen fluid had a downward trend in thepoisoned groups when compared with the control group (P>0.05); ammoniacal nitrogencontent, bacterioprotein, parasite protein and ciliates number were decreased significantly(P<0.05or P<0.01); total acidity was increased significantly (P<0.05or P<0.01). Andwith the increase of concentrations of molybdenum and cadmium, changing trends wereincreasingly obvious.3. When compared with the control group, the pathological anatomy symptoms andthe degree of histologic lesions were increasingly obvious in the poisoned groups alongwith the time of poisoning and the increase of concentrations of molybdenum and cadmium; by means of heart and lung ultrastructure observation, we found the cell nucleuswas deformative, margination and has a structural damage in the high-dose cadmiumcombined with high-dose molybdenum group; the mitochondria was swollen, deformativeand has a fuzzy structure.4. As time went on, the expression of Bcl-2gene was decreased significantly in goats’liver of poisoned groups when compared with control group (P<0.05or P<0.01);expressions of Bax gene and XOD gene were increased significantly (P<0.05or P<0.01);and to some extent, there were some correlation along with the increase of concentrationsof molybdenum and cadmium.The results showed that molybdenum and cadmium irrigating could cause a decline ingoat’s serum free radical, disorder of goat’s ruminal environment, expression change ofapoptosis related gene. Furthermore, with the time of poisoning and the increase ofconcentrations of molybdenum and cadmium, changing trends were increasingly obvious.This indicated that under the molybdenosis and cadmium poisoning in goat, cadmium andmolybdenum had a synergetic effect on serum free radical, ruminal environment andexpression of liver apoptosis ralated gene.
