Zanthoxylunm bungeanum is a kind of deciduous shrub, belongs to Rutaceae. It has greatedibleness and medicinal value. Alkaloids are a class of secondary metabolites in plants whichhave therapeutic efficacies such as anti-tumor, analgesia and antibiotic. However,development and use of Zanthoxylunm bungeanum was limited due to little study on alkaloidin plants of Xanthoxylum. In this investigation, by analyzing the alkaloid content, theextraction process was established and content difference between different pericarps ofZanthoxylunm bungeanum collected in different periods were compared.1. The finally optimized extraction process was as follows: powder of Zanthoxylunm bungeanum wasfiltered through a70mesh sieve and extracted by ultrasonic concussion for30min with70%ethanol asdissolvent. Then heating reflux was operated twice, with each time for2h. In this study, rutaecarpin,berberrubine and ethoxychelerythrine from the pericarp of Zanthoxylunm bungeanum were detected for thefirst time and their HPLC determination method was established as follows: chromatographic column:Agilent ZORBAX SB-C18(4.6mm×250mm,5μm); mobile phase: A:acetonitrile, B: water(with0.1%formic acid); gradient elute:0min30%A,100min80%A; volume flow rate1mL min-1; detectionwavelength260nm(rutaecarpine),265nm(berberrubine),272nm(ethoxychelerythrine); columntemperature:40℃; automatic sampler, sample size:10μL.2. The total alkaloids content of Zanthoxylunm bungeanum from different germplasm resources weresignificantly different. Hanchengdahongpao has the highest total alkaloids content (2.951mg g-1), thenfollowed by Fengxiandahongpao, the Qin’anyihao and Zanthoxylum simulans, respectively. The content ofrutaecarpine, berberrubine and ethoxychelerythrine in different germplasm pericarps has significantdifference. Fengxiandahongpao has the highest content of rutaecarpine and berberrubine, respectively1.184mg·g-1and1.184mg·g-1. The content of rutaecarpine of other germplasm sources was in the followingorder: Zanthoxylum simulans> Hanchengdahongpao> the Qin’anyihao. As for the berberrubine was inthe following order: the Qin’anyihao> Hanchengdahongpao> Zanthoxylum simulans.Hanchengdahongpao has the highst content of ethoxychelerythrine, other provenances was in thefollowing order: Zanthoxylum simulans> Fengxiandahongpao> the Qin’anyihao. 3. The total alkaloids content of Zanthoxylunm bungeanum displayed high level in fruitsetting stage butcontinuously decreased as the fruits growing. During the fruit development, the content of rutaecarpinefirstly increased, then decreased and finally increased until fruit ripening. However, the content ofberberrubine and ethoxychelerythrine increased first, then decreased, and finally achieved the maximumlevel in the early and middle of June repectively.
