Phytosterol was an innoxious natural substance with high physiological activity. Studies find that it has some significant functions, such as anticancer properties, inhibition of tumor, beauty shin. It has been used in steroid drug material, medicine, food and cosmetics widely.Basal part of Phullostachys Pubescens bamboo shoots was the main research object of this paper, which studied the related aspects as follows:nutrient components, type and content of secondary metabolism, test and separation methods of β-sitosterol. At the same time, the parameters of solvent extraction and silica gelcolumn chromatography were optimized, the optimal process parameters were selected to obtain the pure β-sitosterol. The main object of this study was to establish the theoretical basis of bamboo shoots. Results summarized as following:The nutritive indexes of100g dry raw material, such as ash content, crude protein, soluble sugar, dietary fiber were determined, the content of each component was21.71g,5.98g,19.24g,2.98g,17.42g. It follows that basal part of Phullostachys Pubescens bamboo shoots has the characteristics of high protein, high fiber, low fat, etc.The content of allochtonic formations in the raw material was studied, the result indicated that flavonoids content was24.71mg/100g, polyphenols content was30.12mg/100g, triterpenoid content was85.65mg/100g, phytosterol content was245.14mg/100g.This paper made a detailed comparison of Liebermann-Burchard colorimetric method and HPLC methods, the results proved that HPLC method was better for measuring β-sitosterol content: LC-10A tape HPLC instrument, PDA detector, Inertsil ODS-SP,5μm×4.6mm×150mm type chromatographic column, UV detection wavelength:205nm, methanol phase flow, sampling amount:10μL, flowrate:1mL/min, time:25mins. This research measured the β-sitosterol content was230.25mg/100g.By optimization of single factor, Box-Behnken central composite experiment and response surface analysis, solvent extraction was suitable for collecting β-sitosterol, the best conditions were:time:20mins; liquor strength:95%; temperature:55℃; material to liquid ratio:15:1.β-sitosterol was detached and purified for the sake of estimating absorptivity by silica gelcolumn chromatography, the best conditions were:adsorption time:2h; temperature:30℃, concentration of sample:0.3mg/mL, eluent was ethyl acetate: petroleum ether=1:10. The purity analysis of β-sitosterol was80.58%in these conditions. It was found that β-sitosterol purity was enhanced to90.86%through silica gel separation again. The results showed that HPLC, GC-MS and FTIR methods were valuable for reflecting the structure and composition characteristic of β-sitosterol.
