Avian meat quality traits were regulated by genetic and environmental factors and the genetic factors are major. Guangxi sanhuang chicken is one of excellent local species in Guangxi province and has a good meat quality. Therefore, the objective of current study is to analyze the sequence and expression profiles of PPARy and UCP3in different tissues, genders, ages in Guangxi sanhuang chicken and Avian broiler. The correlation was analyzed between the expression levels of PPARγ and UCP3and body weight, abdominal fat, rate of abdominal fat, intermuscular fat. Our result will reveal the molecular mechanism of meat quality and promote the progress of breeding in Guangxi local avian species.Specific primers were designed according to the sequences of PPARγ(AF163811) and UCP3(AF287144) released in GenBank. The target genes were cloned. The result showed that the full sequence of PPARy and UCP3were1428bp and921bp, respectively. There were four base mutations (C291T、G675T、C942T、C1119T) in PPARy and three base mutations (T363G、T399C、C666T) in UCP3in Guangxi sanhuang chicken compared with reference sequences. The mutations in PPARy and UCP3were all synonymous mutations and no change any amino acids. The expression profiles were analyzed using real time PCR in different ages, genders, and tissues in Guangxi sanhuang chicken and Avian broiler. The result showed that expression levels of PPARy were the highest in abdominal fat, the second in subcutaneous fat, a little of in heart, liver, chest muscle and leg muscle. There were very significant differences in species and genders (P<0.01). While expression level of UCP3were the highest in chest muscle, the second in leg muscle, a little of in heart, liver, abdominal fat, and subcutaneous fat. There were very significant difference in species and genders (P<0.01). At the same age, expression levels of PPARy were very significantly higher in females than that in males (P<0.01), while expression profiles of UCP3were very significantly lower in females than that in males (P<0.01). The expression levels of PPARy and UCP3were higher in Guangxi sanhuang chicken than that in Avian broiler at the same age or in the same gender or in the same tissue. The result of correlation analysis indicated that transcription levels of PPARy and UCP3were positive correlation with weight of abdominal fat and intermuscular fat, respectively (R2>0.7, P<0.01).
