The research took Kirin Chicken and Huaixiang chicken as the research materias,determined and compared meat quality,fatty acid and muscle histology characteristics and IMP of the male and the female at 12 、 14 、 16-week old;The single nucleotide polymorphisms of GPAT gene were detected by direct sequencing method and PCR-SSCP methods;The relative expression of green eggshell candidate gene GPAT in heart, liver, breast and leg muscle from Kirin Chicken and Huaixiang chicken in12、14、16-week old were determined by using real-time quantitative PCR(RT-PCR). It aims at group level, individual level and gene level three aspects of the inheritance of IMP content were discussed, and that will provide scientific reference mark the content of broiler IMP.The results showed that:(1) Fatty and muscular histological characteristic. The fat volume of the12 week old male Kirin Chicken were higher than the male the 12 week old fema Huaixiang Chicken(P<0.05);The fat volume of the14 week old female Huaixiang Chicken were significantly higher than the male the 14 week old female Kirin Chicken(P<0.01); The breast muscle fiber density of Kirin Chicken were higher than Huaixiang Chicken(P<0.05).The leg muscle fiber density and breast muscle fiber diameters of Huaixiang Chicken were significantly higher than Kirin Chicken(P<0.05/P<0.01)(2) The IMP content in breast muscle of the 12、16 week old Huaixiang chicken were higher than the 12、16 week old Kirin Chicken(P<0.05); The IMP content in leg muscle of the 16 week old Huaixiang chicken were higher than the 12 week old(P<0.05), were significantly higher than the 14 week old(P<0.01).The IMP content in breast muscle of the 16 week old Kirin chicken were higher than the 12、14 week old(P<0.05). The IMP content in leg muscle of the 14 week old Kirin chicken were higher than the 12 week old(P<0.05), were significantly higher than the 16 week old(P<0.01).(3) SNP.It existed one polymorphic site in Kirin chicken 、Huaixiang chicken、Princess chicken GPAT gene exon 4( G34993A) sites. In the statistic population of Kirin chicken、Huaixiang chicken、Princess chicken, the SNP locus were in the HardyWeinberg non-equilibrium state(P<0.01). Its population genetic structure is stable. PIC groups are moderate polymorphism, It describes Chicken breeds has a wealth of genetic information, certain genetic diversity and breeding potential.(4)GPAT gene expression in Kirin chicken organizations with the increase of age showed decreased trend, GPAT gene expression in Huaixiang chicken organizations with the increase of age showed increased first and then decreased change; GPAT gene Kirin chicken and Huaixiang chicken highest expression in the liver of the three time points;GPAT expression of liver and leg IMP content was negatively correlated. This may be related to differences in species, since the unicorn rolled chicken plumage that has good heat resistance, can reduce the heat stress on visceral injury.
