Pleurotus citrinopileatus Sing, belonging to basidiomycotina, is a precious Fungusand has very high nutritional value and many kinds of medicinal functions. Wild straingrows mainly in the northeast of China while artificially cultivates in the winter of thenorthern areas. It has been developing rapidly in recent years and capable of all-yearcultivation. However, with the increasing demand for Pleurotus citrinopileatus Sing, it hasreceived the consistent praise of consumers, especially in Shanghai, Nanjing, Guangzhouand other southern city, which leading to the rising in prices. The entity of Pleurotuscitrinopileatus Sing can get through artificial cultivation, which has the shortcomings oflong cycle and high cost.Now in contrast of artificial cultivation, industrialized production can overcame thesedefects by means of using liquid fermentation technology. Moreover, the limitation growthin the northern region of China fails to meet the demands of markets in the southern city,and that requires a lot of manpower and financial resources during transportation fromnorth to south.In view of this, here, we study the liquid fermentation technology of Pleurotuscitrinopileatus Sing, including the selection of liquid culture medium and the optimizationof fermentation conditions. In addition, we investigate the conditions and technologiesrequired during cultivation in the middle and lower reaches of the Yangtze Riverregion.The main research content was as follows:1. The liquid fermentation culture medium of Pleurotus citrinopileatus Sing wasscreened by single factor test and orthogonal test and the final optimum flask culturemedium formula was listed as follows: Glucose1.00%, Sucrose1.00%, Peptone0.3%,KH2PO40.15%, NaCl0.03%, MgSO40.07%, VB10.001%.2. To optimize the liquid culture conditions of Pleurotus citrinopileatus Sing in shakeflask, several factors, including the initial pH, fluid amount, quantity, loading speed andtemperature were chosen as indices by means of the combination of single factorexperiment and response surface method. A quadratic polynomial mathematical modelwith good correlation was constructed and employed to the optimization. Results showedthat the optimum culture conditions were as below: Initial pH6.55,Quantity of5%, Fluidamount150mL/250mL, Temperature27℃, Rotational speed155rpm.3. The fermentation of Pleurotus citrinopileatus Sing in shake flask was determinedby the combination of the parameters of liquid shaking bottle quality change and the typical microbial growth curve. Results showed that the fermentation is most appropriatein6~7days.4. Test amplification on the fermenter with7L capacity gived the mycelial biomass9.15g/L, which is conformed well to the result of shake flask test.5. Inoculated strains using strains of solid and liquid, respectively and differentcomposts formula was associated with cultivation methods for cultivation. Besides, weinvestigate the appropriate conditions in the process of spore germination, fruiting andharvest. Compared with solid strains, the results indicate that liquid strains with orderlymycelial age, bigger fruiting body, shorter time in spore germination and fruiting, caneffectively shorten the production cycle and improve production.
