| Our study focused on the enrichment amount of Cry1Ab protein in the Bt rice--Nilaparvata lugens(Stal)--Pardosa pseudoannulata spiders, and investigated the relationship between the Bt enrichment and the activity of the relative enzymes in these spiders. And our results showed:1. The enrichment of Cry1Ab protein existed in the planthoppers fed with the Bt rice, and reached the peak at the10th-15th day. The enrichment of Cry1Ab protein existed in the spiders which fed with planthoppers eating Bt rice10th-15th day, and reached the peak at the10th day; besides, the Bt contents in the3th instar of Pardosa Pseudoannulata was higher than that of5th instar.2. The CrylAb protein existed in digestive system tissue, muscle tissue (appendages) and blood lymphocytes of adult spiders, instead of nerve tissue; and the Cry1Ab protein in mitochondria and microsome of spiders were higher than that of cell debris or cytoplasmic extracts; CrylAb protein was also detected in mitochondria and microsome, cell debris, cytoplasmic extracts in the3th instar and5th instar of Pardosa Pseudoannulata.3. Only AchE activities in muscle tissue and nerve tissue of experimental male spiders, muscle tissue of experimental female spiders and SOD activities in muscle tissue of experimental male5th spiders were significant higher than the control groups (p<0.05); in different maturity, AchE activities in digestive system of experimental3th instar spiders, POD activities in muscle tissue of experimental5th instar spiders were significantly higher than the control (p<0.05). However, activities of other enzymes in all tissue had no significant difference compared with the control groups (p<0.05). It showed that the enrichment of CrylAb protein had little effects on enzymes activities in spiders.4. Only AchE activities in mitochondria and microsome of digestive system tissue, nerve tissue, blood lymphocytes of experimental male spiders, in cytoplasmic extracts of muscle tissue, blood lymphocytes of experimental male spiders, in cytoplasmic extracts of muscle tissue, blood lymphocytes of experimental female spiders had significant difference with the control groups (p<0.05); SOD activities in cell debris of muscle tissue of experimental male spiders, CAT activities in mitochondria and microsome of digestive system of experimental female and male spiders, POD activities in mitochondria and microsome of blood lymphocytes of experimental male spiders and in cytoplasmic extracts of muscle tissue of experimental female spiders had significant difference with the control groups (p<0.05); activities of others enzymes in all subcellular structure had no significance with the control groups. While in different instar spiders, POD activities in cytoplasmic extracts of digestive system and muscle tissue of experimental3th instar spiders were significantly higher than the control groups (p<0.05). POD activities in cytoplasmic extracts of muscle tissue of experimental5th instar spiders were significantly higher than the control groups (p<0.01); GSH-Px activities in cytoplasmic extracts of blood lymphocytes of experimental3th instar spiders were significantly higher than the control groups (p<0.05). AchE activities in mitochondria and microsome of muscle tissue of experimental3th instar spiders were significantly lower than the control groups (p<0.05); but in nerve tissue of experimental3th instar spiders were significantly higher than the control groups (p<0.05). AchE activities in cytoplasmic extracts of digestive system of experimental3th instar spiders were significantly higher than the control groups (p<0.05). However, activities of other enzymes in all subcellular structure had no significance with the control groups.In our results, CrylAb protein could bioaccumulate in muscle tissue from digestive system tissue to blood lymphocytes. All the Bt enrichment has some effects on the enzyme activities, but whether it affects the spiders’ safety needs further study. |
