Objective To the expression of Cdk5protein in gastric cancer andresearch its association with the clinicopathologic features,andinvestigate its influence in carcinogenesis of gastric mucosa andprognosis.Methods206cases of gastric cancer tissues,54cases of adjacentnon-cancerous tissues and34cases of lymph node metastatic cancer tissuesparaffin-embedded specimens were collected from the Union Hospitalaffiliated to FuJian Medical University with detailedclinicopathological parameters in December2006to September2007;Immunohistochemical SP method was used to detect the expression level ofCdk5.16cases of gastric cancer tissues and its’adjacent non-canceroustissues were collected after operation in May to June of2012; Westernblot assay was used to detect the expression level of Cdk5. The associationof Cdk5expression with clinicopathological parameters was analyzed bySPSS17.0software.Results Results1.Cdk5protein can express in gastric cancer tissues,adjacent non-cancerous tissues and lymph node metastatic cancer tissues,and mainly in cytoplasm. The rate of Cdk5protein expression in gastriccancer tissues was considerably lower than that in adjacent non-canceroustissues (P<0.05). The rate of Cdk5protein expression in lymph nodemetastatic cancer tissues was considerably lower than that in gastriccancer tissues.2. The lower expression of Cdk5protein in gastric cancerwas associated with Borrmann type, grade of differentiation, depth ofinvasion, lymphatic metastasis and pTNM stage. Loss of Cdk5proteinexpression had a significant correlation with poor survival both in univariate and multivariate analyses.3. Detected by Western blotanalysis software, Cdk5protein relative expression intensity in gastriccancer tissues (0.4355±0.4185) was significantly lower than its adjacentnon-cancerous tissues (0.7962±0.4945), the difference was statisticlysignificant (P<0.05).Conclusion These results indicate that Cdk5protein was down-regulatedin gastric cancer, and the abnormal expression of Cdk5protein may berelated to carcinogenesis and the development of gastric cancer....
