Objective:To observe the effect of Sulfortanshinone sodium injection(STS) in guinea pigs withacute lung injury,we injected different concentrations of Sulfortanshinone sodium injection(STS) toguinea pigs with acute lung injury induced by firearm wound.Methods:60guinea pigs wererandomized to six groups: normal control group(N)、acute lung injury model group(A)、physiological saline injection in control group(P)、and low dose of STS treatment group(L), middledose of STS treatment group(M),high dose of STS treatment group(H),every group has ten guineapigs. First, we record the weight of every guinea pig. We used10%hydration chlorine was used tomake guinea pigs of group A, P,L,M and H into anesthesia, and then put the guinea pigs into hightemperature gas experiment device platform to inhaled150℃high temperature air about3minutes, which set up acute lung injury model. After half an hour, the group L guinea pigs wastreated with the STS10mg/kg, and the group M guinea pigs was treated with the STS20mg/kg,andthe group H guinea pigs was treated with the STS30mg/kg, which they were observed the breathand activities of guinea pigs. After12hours, we have obtained blood of groups of guinea pigs fromthe orbital venous.Guinea pigs were made into anesthesia again and then it was extracted2mlblood from abdominal aortic for arterial blood gas analysis, there were2ml blood which wasextracted from abdominal aortic venous were used to detect the concentration ofelastase,interleukin-8,TNF alpha and NF-κB in serum. The pigs were killed, and which they wereexrtacted from the right lung tissue to calculate wet/dry weight (W/D) ratio, the upper left lungtissue was also taken away to dye with HE and then to observe the pathological changes of lungusing naked eye and light-microscopy.Results1.Analysis of arterial blood gas:Group A,P,L,M,Hare significantly lower than group N in PaO2and SaO2(P<0.05), higher than N group inPaCO2(P<0.05); Group M are siginificantly lower than group N in PaO2and SaO2(P<0.05),but higher than group A(P<0.05);Group M are lower than group A(P<0.05),but closed to groupN(P>0.05).2.Lung wet and dry weight (W/D)ratio: In lung wet and dry weight,group A,L,M,H arehigher than group N(P<0.01).Group P has no difference with group A(P>0.05).Group L,M,H arelower than than group A(P<0.01).Group M has difference with group L(P<0.01),but group H has nodiference with group L(P>0.05).Group M has difference with group H(P<0.01)3.Lung tissuemorphology:No obvious injury, the complete alveolar structure, no alveolar walls edema andinterstitial pulmonary infiltrated by a few leucocyte were detected in group N. We have found themost obvious lung injury in group A,P,L,H, such as there are the pulmonary interstitial edema、alveolar edema、exudate volume and a lot of red cell in lung of the pigs, which were found alveolarinterstitial thickening, telangiectasia, congestion,widened alveolar septum, there were manyinflammatory cells infiltration in group M, which is light damage,and closed to group N.4.Cytokinelevels:The cytokine of tumor necrosis factor alpha、interleukin-8and nuclear factor-κB level ingroup A,P,L,M,H are obviously higher than the group N(P<0.01).After treatment of STS,thecytokine of tumor necrosis factor alpha、interleukin-8and nuclear factor-κB levelgroup L,M,H arelower than group A(P<0.05).And group M decreased the most obvious, which was closed to groupN(P>0.05).Conclusion:1.It is typical and reliable to make the model of ALI by guinea pig throughfirearm.2.The lung tissue injury can be effectively protected by the STS.With the increase of STSdosage,they were obviously plaied role in repair of the pigs with lung injury.And after the treatmentof the middle dose of STS, it was the strongest effect in their lung tissue protection. While thesaline treatment had no effect in ALI, but it can be distinguished as compared with the effect ofSTS.
