Objective:Asthma is by a verity of cells,especially mast cell,eosinophils,T lymphocytes,neutrophils,airwayepithelial cell and a varity of cellular components in the chronic airway inflammation disease ofairway inflammation and the reactive as the main feature. This experiment through the establishmentof animal model of asthma,and to give vitamin K1intervention, compare cell count andclassification in the rat bronchoalveolar lavage fluid (BALF),the level of IL-4in BALF and lungtissue expression of ICAM–1.This paper discusses the vitamin K1in asthma of anti-inflammatoryeffects and its possible mechanismMethods:In accordance with the principles of randomization,30male SD rats were randomlydivided into three groups,asthma group,vitamin K1and control group(n=10).Rats ofasthmatic group and vitamin K1group were immunized on day1,day8and day15byintraperitioneal injection of100mg ovalbumin(OVA) in1ml of saline with100mg ofaluminum hydroxide. From day16these animals were challenged with aerosolizedOVA (1%in saline) for30minutes per day for7consecutive days. From the16thday,OVA sensitized rats of vitamin K1group were given intraperitoneal injection ofvitamin K10.5mg/(kg d) per day for7consecutive days. OVA and vitamin K1replaced by normal saline (NS) in control group. In the24hours after the last challenge,rats were killed to collect lung tissue and airway lavage fluid (BALF). BALF cells sandsupernatant were separated by centrifugal separation. Iung tissue was made into lung biopsy specimens through dehydration and paraffin embedding. Total cells anddifferentiated leukocytes in BALF were counted through the improved cattle’s counttable. Interleukin4(IL-4) levels in BALF supernatant were measured by elisa method.Intercellular adhesion molecule1(ICAM-1) levels of lung specimens were assessedwith the use of immunohistochemical. Each group of indicators data were analyzed andcompared by statistical methods.Results:In the asthma group, the count of acidophilus granulocyte and neutrophilegranulocyte、the level of IL-4in BALF and the expression of ICAM-1in lung tissues,(7.10±2.89)×10~6vs(0.80±0.79)×10~6,(31.30±6.00)×10~6vs(7.40±1.96)×10~6,(615.37±94.77)pg/ml vs(470.39±20.51)pg/ml,(0.34±0.10)vs(0.17±0.04),hadsignificant differences compared with those of in the control group, respectively(P<0.05). The count of acidophilus granulocyte(4.60±1.70)×10~6and neutrophilegranulocyte (21.20±6.19)×10~6and the level of IL-4(542.40±29.17)pg/ml in BALFand the expression of ICAM-1(0.25±0.04)in lung tissues were significantly lowerthan those in the asthmatic group.Conclusions:Vitamin K1can significantly decreasethe expression of the level of IL-4in BLAF and ICAM-1in lung tissues,thus alleviatesairway inflammation.Conclusion:Vitamin K1can significantly decrease the expression of the level of IL-4in BLAF andICAM-1in lung tissues,and alleviates airway inflammation of asthmatic mice as well....
