OBJECTIVE To study the mechanism of WUJIBAIFENGWAN in therapy ofchronic nonbacterial prostatitis. METHODS The model of chronic nonbacterialprostatitis in rats was established by injecting with XIAOZHILING solution into theprostates.The sham-operated rats were dealt with saline instead of XIAOZHILINGsolution into the prostates. The sham-operated rats and chronic nonbacterial prostatitisrats were divided into5groups at first week after surgery,8rats in each group:(1)sham-operated group: rats were given the same amount of saline everyday;(2) CNPmodel group: rats were given the same amount of saline everyday;(3) low-does ofWUJIBAIFENGWAN group: rats were given WUJIBAIFENGWAN (1g·kg-1per day);(4) high-does of WUJIBAIFENGWAN group: rats were given WUJIBAIFENGWAN(2g·kg-1per day);(5) QIANLIETAI group: rats were given QIANLIETAI (0.95g·kg-1per day). The general situations of rats were observed. All groups were intragastricadministered medicine for4weeks after surgery. Then, all rats were killed and theprostates of all rats were observed and weighed. Hematoxylin-eosin staining, massonstaining were used for morphological observation and fibrosis analysis of the prostates.Immunohistochemical staining and Western Blotting analysis were used to observe theexpressions of TGF-β1and Smad4in the prostates. RESULTS (1) General situationof animals: sham-operated rats were full of vigour, have smooth hair and their weightwas added. The weight of CNP model rats was significantly lightened, their hair wasscrawny and their mental state was poor. The general situation was improved aftertreatment.(2) Appearance of prostates: the prostates of the sham-operated rats weresmooth and rosy, the structure was complete and was easily peeled from surroundingtissue. The prostates of the CNP rats were dark red or brown and hard, the structurewas incomplete and adjoining with surrounding tissue. Compared with the CNP ratsprostate, the treated rats prostates have been improved in some degree. Some prostatesof treated rats were similar to those of sham-operated rats.(3) The prostate index: compared with the sham-operated rats, the prostate index of CNP rats increasedobviously (P<0.05); compared with the CNP rats, the prostate index of treated rats wasall decreased (P<0.05).(4) HE staining: the prostatic tissue structure of sham-operatedrats was complete; the prostatic tissue structure of CNP rats was significantlyabnormal; the prostatic inflammatory infiltration of treated group rats had improved.(5) Masson staining: compared with the sham-operated rats, the prostatic fibrousconnective tissue of the CNP rats increased obviously (P<0.01); the degree of fibrosisreduced after treatment (P<0.05).(6) Immunohistochemical staining: the expressionsof TGF-β1and Smad4in the prostate of CNP rats were significantly increased; theexpressions were inhibited after treated with WUJIBAIFENGWAN (P<0.01).(7)Western Blotting: compared with the sham-operated rats, the expressions of TGF-β1and Smad4protein in the prostate of CNP rats were significantly increased (P<0.05);the protein expressions were decreased after treated with WUJIBAIFENGWAN(P<0.05, P<0.01). CONLUSIONS (1) The pathological changes in prostate of rat withchronic nonbacterial prostatitis caused by XIAOZHLING injection are extremelysimilar to that of clinical chronic prostatitis.(2) The TGF-β/Smad signaling pathway isinvolved in the pathophysiological process of CNP.(3) WUJIBAIFENGWAN iseffective to cure chronic nonbacterial prostatitis of rats; its mechanism is probablyrelated with the decreasing production of TGF-β1and Smad4.
