Objective: To explore the relationship of HIF-1α (hypoxia-induciblefactor-1α)、β-catenin and Wnt pathway, and the role of IMP3(insulin-likegrowth factor Ⅱ mRNA—binding protein3)and PCNA (proliferating cellnuclear antigen)on the proliferation of gastric cancer SGC-7901cells.Methods: pcDNA6.2-GW/EmGFP-miR-β-catenin plasmid wastransfected into SGC-7901cells and established stably transfection celllines miR-β-catenin-7901. In this experiment, four groups were designed:control (SGC-7901cells were cultured under normoxic), hypoxia(SGC-7901cells were cultured under hypoxia), interference(miR-β-catenin-7901cells were cultured under normoxic)and hypoxiainterference (miR-β-catenin-7901cells were cultured under hypoxia)groups. The proliferation characteristics of SGC-7901cells in four groupswere detected by colony formation and doubling time assay. Theexpression levels of HIF-1α, β-catenin, IMP3and PCNA proteins ofSGC-7901cells in four groups were detected by Western blotting. Results: As compared with control, the number of colony formationwas increased and doubling time was shorter in hypoxia group (P <0.05).As compared with the hypoxia group, the number of colony formation wasreduced and doubling time was longer in hypoxia interference group (P <0.05). The expression levels of HIF-1α, β-catenin, IMP3and PCNAproteins in hypoxia group were higher than those in the control group (P<0.05). The expression levels of HIF-1α, β-catenin, IMP3and PCNAproteins in interference group compare with control group were lower (P<0.05). The expression of HIF-1α, β-catenin, IMP3and PCNA proteins inhypoxia interference group were lower than those in hypoxia group (P<0.05).Conclusion:1) miR-β-catenin-7901cell line of stability interfered theβ-catenin which was constructed successfully。2) under hypoxic condition,the proliferation of gastric cancer cell line SGC-7901can enhance.3)Under the hypoxic environment, the proliferation of Gastric cancercell may be related to which HIF-1may activate the Wnt pathway throughtbeta-catenin and it raised IMP3and PCNA expression levels...
