At present,in cancer deaths in the world,gastric cancer ranks second which is second only to lung cancer. While in china,gastric cancer is one of the highest morbidity and mortality of malignant tumor.Numerous studies have demonstrated,gene mutation and cell proliferation acceleration is widespread human cells canceration mechanism.8-OHd G is due to Reactive oxygen species against DNAgive rise to guanine eighth hydroxyl. Can measure oxidative stress levels in the cell,and after the metabolic stability there,fluids and related tissues and organs of 8-OHd G level could evaluate the risk of attack.For disease progression and treatment effect is more intuitive and effective information.PCNA is a kind of cell cycle regulatory proteins, it's expression is imparity in different periods of cell cycle, and only in the proliferation of the state of the cell,at its peak in sphase of cell proliferation.Can be roughly think and the quantity of DNA synthesis. So it can be regarded as an index of cell proliferative activity when it was detected by immunohistochemical method and calculated the positive rate of various tissues.Preliminary discussion in both its effect in the process of gastric cancer and correlation provides a special index system for evaluate gastric cancer biological behavior and estimate prognosis of malignancy.Samples of 63 tumor tissues and 36 paracancerous tissues and 9 Chronic Gastrits mucosa of patients undergoing surgery for gastric cancer were analyzed. PCNA protein expression in these specimens was examined by immunohistochemistry.DNA was isolated from these specimens.Then, DNA was hydrolyzed to nucleotides. The 8-OHd G were determined in hydrolysates by high-performance liquid chromatography(HPLC) with UV detector. The correlation between the expression of PCNA protein and the level of 8-OHd G and clinicopathologic parameters of gastric carcinoma was analyzed.The rate of PCNA protein overexpression in gastric carcinoma were 88.9%(56/63),there was statistical significances in the rate of PCNA protein overexpression between gastric carcinoma and paracancerous tissue 60.0%(21/36). Moreover, they were significantly higher than those in Chronic Gastrits mucosa(P<0.05). PCNA protein overexpression was associated with tumor differentiation and TNM stage(76.9%vs97.3%, 77.3%vs95.1%).The levels of 8-OHd G in gastric carcinoma(39.84±13.01?g/m L) were higher than paracancerous tissue(15.95±5.02?g/m L) and Chronic Gastrits mucosa(7.99±2.03?g/m L).8-OHd G levels were associated with tumor differentiation and TNM stage(39.15±13.88?g/m L vs 44.81±9.67?g/m L, 39.56±13.69?g/m L vs 44.01±10.56?g/m L, P<0.05). PCNA protein overexpression is relevould like 8-OHd G levels.No significant differences were shown in 8-OHd G levels and PCNA protein overexpression in those three tissues between age and sex and tumor type or lymph node metastasis.According to above results, up-regulation of PCNA protein expression and 8-OHd G levels was observed in gastric carcinoma. Detection of PCNA protein expression and 8-OHd G levels can be used to evaluate malignant biological behavior and prognosis of gastric cancer.
