Objective:To investigate y the effects of TPP on the reproductive toxicity in male pubertal rats. And the influence of expression of Proliferating Cell Nuclear Antigen and AROMatase in testis, to explore the mechanism of TPP on male reproductive system.Methods:40health21days-old SD rats were divided into four groups randomly:control group, low dose group (250mg/kg),medium dose group (500mg/kg), high dose group (750mg/kg). The treated rats were orally administered various doses of TPP. After30days, sacrificed the rats for experiments. Observe the change of histopathology in testicular by HE stain. Immunohistochemical was used to observe PCNA, AROM protein expression in testicular of each rat. We extracted the PCNA, AROM mRNA on the rat testicular tissue for RT-PCR and using Western blot measure the PCNA,AROM protein expression.Result:The body weight of rats in treatment groups were significantly lower than the control group(P<0.05). But the numbers of spermatogenic cells in testicular tissues were not different in each groups,(P<0.05).HE stain showed the structure of seminiferous tubule is integrity, and seminiferous epithelial cells arranged orderly in control group, while histopathologic examination revealed serious degeneration of seminiferous epithelium, sperm tangles serious in treat groups. Immunohistochemical tests showed the expression of PCNA and AROM reduced obviously in treat groups(P<0.05). RT-PCR results showed that the PCNA, AROM mRNA expression reduced, and it had a significant difference with the control group(P<0.05). Western blot results showed that the PCNA, AROM protein expression reduced,and it had a significant difference with the control group(P<0.05).Conclusion:exposure to TPP can lead to damage of male reproductive system, restrain the PCNA, AROM mRNA and protein expression in testis of rats. It suggests that rat exposure to TPP during the puberty can damage the expression level of PCNA, AROM in the testicular tissues, and damage the testis development in the future.
