Objective: To investigate the effect of decorin gene transfection on thegrowth of nude mice xenograft which bearing human gastric carcinoma cellSGC-7901and learn the possible mechanisms.Methods: Gastric carcinoma xenograft model in nude mice wasestablished, recombinant plasmid PEGFP-N1-DCN was injected to thecenter and bottom of the carcinoma, with the physiological saline groupand empty plasmid group as control groups, the change of carcinomavolume and weight was measured, the expression of DCN was detected byRT-PCR and western blot, the expression of transforming growthfactor-β1(TGF-β1) and Matrix Metalloproteinases-9(MMP-9) protein wasmeasured by western blot, the expression of vascular endothelial growthfactor (VEGF) was assessed by immunohistochemistry.Results: Gastric carcinoma xenograft model in nude mice wasconstructed successfully, compared with the control groups, slower growthspeed, smaller volume and lighter weight was found in recombinantplasmid group; RT-PCR showed that the expression of decorin gene in recombinant plasmid group was significantly higher than control groups,western blot indicated that the expression of fusion protein was found inrecombinant plasmid group; western blot showed that the expression ofTGF-β1and MMP-9protein in recombinant plasmid group was lower thancontrol groups; immunohistochemistry indicated that the expression ofVEGF was also down-regulated in recombinant plasmid group.Conclusion: Decorin gene can inhibit the growth of nude micexenograft, decrease the expression of TGF-β1and MMP-9protein andinhibit the angiogenesis of tumor may be the crucial mechanisms.
