Objective: Fibroblast growth factor19(FGF19) is a novel endocrinehormone that regulate bile acid metabolism, gallbladder refilling,glucose and lipid metabolism. There is little evidence on therelationship between serum FGF19levels and leptin-insulin resistance. Thehuman data of serum FGF19levels in prediabetes is also limited. In orderunderstand the effects of FGF19in the ocurrence and development ofdiabetes, we measured the change of serum FGF19levels in patients withnormal glucose tolerance (NGT),impaired glucose tolerance(IGT)and type2diabetes (T2DM) and evaluated the relationship between FGF19and leptinand other parameters.Methods:30patients with newly diagnosed T2DM,30patients with IGTand30individuals with NGT were involved in this study. All the subjectsincluded52MS ones and38non-MS ones. Fasting serum FGF19and leptinlevels were measured by ELISA. Other parameters were also analyzed bystandard methods.Results: Fasting serum FGF19levels were significantly decreased in T2DM and IGT groups than in NGT group (256.12±61.66vs.312.89±62.57vs.391.84±63.46ng/L,P<0.01). MS subjects had obviously decreased serumFGF19levels than non-MS subjects (284.51±67.91vs.391.84±63.46ng/L,P<0.001). Fasting serum leptin levels showed an opposite trend like FGF19in IGT, T2DM and MS groups. Serum FGF19levels were negativelycorrelated with waist circumferences (WC), fasting insulin (FINS),homeostasis model assessment of insulin resistance (HOMA-IR), leptinlevels in IGT, T2DM, MS groups. Moreover, WC, HOMA-IR, leptin werethe independently predictors of FGF19levels in T2DM groups.Conclusion: Decreased FGF19levels may be a contributing factor in theoccurrence and development of T2DM. Decreased FGF19levels wereclosely related to leptin-insulin resistance,therefore,FGF19might have theopportunity to play a role in T2DM and abdominal obesity control.
