Charpter One Relations between serum leptin and TNF-αand insulin resistance in rat model of the metabolic syndromeObjective:To observe the relations between insulin resistance and leptin and TNF-αin the rat model of metabolic syndrome induced by high fat and high sucrose diet, moreover, to research the roles of leptin and TNF-αin the process of the metabolic syndrome. If they mediate the development of insulin resistance? And as inflammatory factors, if they can be regarded as predictions of cardiovascular diseases.Method: Homa IR is used to estimate the insulin resistance. Serum leptin and TNF-αare detected by RI and ELISA respectively. Leptin in adipocyte is detected by Immunohistological(ABC) method.Result: Compared with control group, weight, fasting blood glucose, fasting plasma lipid and insulin of rats in experimental group are significantly increased(p<0.01). Index of insulin resistance are increased too[(4.44±0.27:5.45±0.51),P<0.05 ], plasma leptin and TNF-αare obviously increased[(0.15±0.05 :0.41±0.23,P<0.01].Conclusion: High fat and high sucrose diet induce the model of rat of metabolic syndrome. There are close relationship between insulin resistance and serum leptin and TNF-α. Leptin may be mediate the produce of insulin resistance. And TNF-α, as the inflammatory factors , can predict the cardiovascular diseases. Charpter Two Effect of rosiglitazone on leptin and TNF-αof the metabolic syndrome ratsObjective To study the effect of rosiglitazone on leptin and TNF-αof the metabolic syndrome rats by observating the change of leptin and TNF-α.Method: Rats of the same age were divided into 3groups randomly: the normal control group, fat group and the rosiglitazone-treated fat group . Homa IR is used to estimate the insulin resistance. Serum leptin and TNF-αare detected by RI and ELISA respectively. Leptin in adipocyte is detected by Immunohistological(ABC) method.Results: After 4 weeks, Leptin and TNF-αof the rosiglitazone-treated fat group were changed obviously(0.41±0.23: 0.19±0.49,p<0.05)( 356.83±87.05: 196.92±32.48,p<0.01).Conclusion: High fat and high sucrose diet induce the model of rat of metabolic syndrome. There are close relationship between insulin resistance and serum leptin and TNF-α. Function of rosiglitazone is implemented by effection on leptin and TNF-α..
