PART ONE: Expression and significance of Yes-associated protein inprostate cancerObjective: To investigate the expression and clinical significance ofYAP in the tissue of prostate cancer (PCa) tissue, pericarcinoma (PC) tissueand benign prostate hyperplasia (BPH) tissue.Methods: The expression of YAP protein of32cases of PCa,15casesof PC and15cases of BPH was investigated by immunohistochemistry.Moreover; the different expression of YAP protein of15cases of PCa andits corresponding PC tissue was confirmed by western-blot.Results: The rate of YAP positive staining (78.13%) in PCa washigher than that in PC tissue (26.67%), and the YAP positive staining wasnot seen in the BPH tissues. The staining difference between each groupwas statistically significant (P=0.000). Furthermore, the differentexpression of YAP protein was relevant to clinical stage and the degree of differentiation (P=0.019, P=0.012). The statistically significant differencesof YAP protein expression were observed in15cases of PCa compared toits corresponding PC tissue by western-blot (P=0.006).Conclusions: The expression of YAP protein was significantly higherin PCa compared to non-tumorous tissues. Expression of YAP shows theobvious tumor specificity in PCa. YAP may be a new target in the diagnosisand treatment of the PCa. PART TWO:The research of interaction mechanism between YAP andAR and its role in CRPC transformingObjective:To investigate the expression of YAP in prostate cancercells PC3, LNCaP, DU145and benign prostatic hyperplasia cells BPH-1,choose hormone-dependent prostate cancer cells LNCaP to make apreliminary exploration of the interaction between YAP and AR and its rolein CRPC transforming.Methods: The expression of YAP protein in prostate cancer cells PC3,LNCaP, DU145and benign prostatic hyperplasia cells BPH-1wasconfirmed by western-blot. Co-immunoprecipitation technique was adoptedin hormone-dependent prostate cancer cells LNCaP using YAP proteinprecipitation AR. The gene transfection technology was used to silence YAP in LN-Cap cells and the expression of YAP and AR was detected bywestern-blot.Results: The expression of YAP protein was significantly higher inthree kinds of prostate cancer cells than the hyperplasia of prostate celllines BPH-1, and hormone dependent cell line LNCaP was higher than thehormone independent cell lines PC3and DU145.YAP proteins canco-immunoprecipitation AR in LNCaP cells, the AR expression wasdecreased obviously when YAP gene was silent with siRNA in LNCaP cellline.Conclusions: The expression of YAP protein was significantly higherin prostate cancer cells compared to benign prostatic hyperplasia cells, theinteraction between YAP and AR may play an important role in CRPCtransforming, which is necessary to deeply investigate.
