Object:To investigate the expression of PTEN, PI3K of PTEN/PI3K/AKT signaling pathway in colon carcinoma tissue and adjacent normal tissue, analyzing the correlation between its expression and the clinicopathological factors、the relationship of their expression in colon carcinoma to explore its role in the development and prognosis in colon carcinoma.Methods:We randomly selected40patients with adenocarcinoma of colon cancer pathologically confirmed after operation in First Affiliated Hospital of Dalian Medical University from January2005to May2008.All patients had no chemotherapy and other antitumor therapy, including23males and17females, aged31years old to88years old, with a median age of50.5years;3cases of well-differentiated,28cases of moderately differentiated,9cases of the poorly differentiated; lymph node metastasis in9cases, without lymph node metastasis in31cases;3cases of stage Dukes A,28patients with stage Dukes B,9patients with stage Dukes C; The clinical data of the patients is integrity.Follow-up was the date of surgery to the date of death or last follow-up (60months). In another40cases from the tumor margin5cm outside the normal colonic mucosa tissue as a normal control group.2. Using Immunohistochemistry to detect the expression of PTEN, PI3K in40cases of colon carcinoma tissue and40cases of colon tissue.3. SPSS11.5statistical software was used to test the significance and relevance. Using X2test and Fisher’s exact test to analyze the relationship between the positive rate of PTEN、PI3K and clinical pathological factors、five-year survival rate; Using Spearman for analyzing the relationship between the expression of PTEN and PI3K in colon carcinoma. Significance level a=0.05(two-tailed), P<0.05means that there is a significant difference.Result:1、There were21cases of PTEN positive expression in40colon carcinoma tissue,the positive rate of PTEN was52.5%,meanwhile, there were38case of PTEN positive expression in40normal colon tissue,the positive rate of PTEN was95.0%,there was significantly difference between colon carcinoma tissue and normal colon tissue(X2=10.838, P<0.05).The expression of PTEN was not associated with sex,age,tumor location,size(P>0.05),which is closely related to the degree of differentiation,depth of invasion,tumor stage,lymph node metastasis.In moderately differentiated group,the positive rate of PTEN was64.52%, in the low differentiated group,the positive rate of PTEN was11.11%,there was significantly difference in two groups(X2=7.977,P<0.05);in the infiltration≤T2group,the positive rate of PTEN was85.71%,and in another group,the positive rate of PTEN was45.45%, there was significantly difference in two groups(X2=3.754, P<0.05);In the clinical stage of Dukes A and B group,the positive rate of PTEN was61.29%,while the positive rate of PTEN wan22.22%in Dukes C group,there was significantly difference in two groups(X2=4.269, P<0.05);The positive rate of PTEN was22.22%in lymph node metastasis group,however,it was61.29%in the group without lymph node metastasis, there was significantly difference in two groups(X2=4.269, P<0.05).2、In40cases of colon carcinoma patients, the survival of more than5years were30cases, of which the positive rate of PTEN was60.00%, while in the10patients with a survival time of less than5years, the positive rate was30.00%, the differences between the two groups was statistically significant (X2=4.269, P<0.05).3、There were28cases of PI3K positive expression in40colon carcinoma tissue,the positive rate of PI3K was70.00%,meanwhile, there were4case of PI3K positive expression in40normal colon tissue,the positive rate of PI3K was10.00%,there was significantly difference between colon carcinoma tissue and normal colon tissue(X2=13.714,P<0.05),the expression of PI3K was not correlation with sex,age,tumor location,size,degree of differentiation,depth of invasion,tumor stage,lymph node.4、In40cases of colon carcinoma patients, the survival of more than5years were30cases, of which the positive rate of PI3K was66.67%, while in the10patients with a survival time of less than5years, the positive rate was80.00%, the differences between the groups was statistically significant (X2=4.269, P<0.05).5、Analyzing the expression of PTEN and PI3K in40cases of colon carcinoma tissue,there were13cases which the two proteins were positive;there were2cases which the two proteins were negative;there were10cases which the expression of PTEN was positive and the PI3K was negative;there were16cases which the expression of PTEN was negative and the PI3K was positive; The expression of PTEN was significant negative correlation with the expression of PI3K by the Spearman (r=-0.332, P<0.05)Conclusion:1. PTEN involves in the occurrence of colon cancer, and relates to colon cancer invasion, metastasis-associated.It may become a predictor of prognosis of colon cancer.2. PI3K is associated with colon carcinoma, but it is not related to the invasion of colon cancer metastasis and prognosis.3. The lack expression of PTEN in colon cancer may activate the PI3K signaling pathway, both jointly involve in colon cancer and they may provide the basis for individualized treatment for colon cancer.
