The EliVisionTM Plus immunohistochemical technique and in situ hybridization technique was used to detect Snail, E-cadherin, VEGF protein and mRNA in laryngeal mucous membrane chronic inflammation, atypical hyperplasia and squamous cell carcinoma tissues.To search the expression of Snail, E-cadherin, VEGF protein and mRNA in laryngeal mucous membrane chronic inflammation, atypical hyperplasia and squamous cell carcinoma tissues and their relations to clinical pathological features. To detect the relations among Snail, E-cadherin, VEGF protein and mRNA in laryngeal squamous cell carcinoma tissues. As well as the consistency of the expression of Snail, E-cadherin and VEGF using immunohistochemisty and in situ hybridization.Sixty cases of laryngeal squamous cell carcinoma,30cases of atypical hyperplasia and20cases of chronic inflammation tissues were collected from the department of pathology in the251st hospital of PLA ranging from January in2005to January2007. The expression of Snail, E-cadherin, VEGF protein and mRNA were detected in three tissues using immunohistochemistry technique of EliVisionTM Plus and in situ hybridization technique. To analyse the relations among the three markers and the gender, age, pathology classification, TNM stage, T stage, thyroid cartilage infringe upon, lymph node metastasis and distant metastasis. The consistency of the expression of three tissues by using immunohistochemisty and in situ hybridization was analyzed.The positive rate of Snail, E-cadherin, VEGF protein and mRNA in60cases Laryngeal Squamous cell carcinoma tissues was78.3%(47/60), 46.7%(28/60),80.0%(48/60) and70.0%(42/60),45.0%(27/60),73.3%(44/60); The positive rate of Snail, E-cadherin, VEGF protein and mRNA in30cases atypical hyperplasia tissues was43.3%(13/30),76.7%(23/30),36.7%(11/30) and20.0%(6/30),70.0%(21/30),26.7%(8/30); The positive rate of Snail, E-cadherin, VEGF protein and mRNA in20chronic inflammation tissue was15.0%(3/20),100%(20/20),15.0%(3/20) and5.0%(1/20),90.0%(18/20),5.0%(1/20). The positive rate of Snail, VEGF protein and mRNA in laryngeal squamous cell carcinoma tissues was significantly higher than that in atypical hyperplasia and chronic inflammation tissues; The positive rate of E-cadherin protein and mRNA, in laryngeal squamous cell carcinoma tissues was significantly lower than that in atypical hyperplasia and chronic inflammation tissues, and it was statistical significance. The positive rate of Snail protein in atypical hyperplasia tissue was higher than that in chronic inflammation tissues, and it was statistical significance; The positive rate of Snail mRNA, VEGF protein and mRNA was higher than that in chronic inflammation tissues, but it was not statistical significance. The positive rate of E-cadherin protein in atypical hyperplasia tissue was lower than that in chronic inflammation tissues, and it was statistical significance; The positive rate of E-cadherin mRNA in atypical hyperplasia tissue was lower than that in chronic inflammation tissues, but it was not statistical significance. The overexpression of Snail, VEGF protein and mRNA and the lower expression E-cadherin protein and mRNA were correlate to the TNM stage, T stage, thyroid cartilage infringe upon, lymph node metastasis, distant metastasis and5year survival rate, while not correlated to age, gender. The expression of Snail protein was correlate to pathology classification, while the expression of Snail mRNA was not correlate to it.The positive cases of Snail, E-cadherin, VEGF protein and mRNA in the60larynal squamous cell carcinoma cases were47,2,848and42,27,44respectively. Negative correlation was found through spearman test between the expression of Snail protein and mRNA and E-cadherin protein and mRNA; Positive correlation was found between the expression of Snail protein and mRNA and VEGF protein and mRNA; Negative correlation was found between the expression of VEGF protein and mRNA and E-cadherin protein and mRNA.The overexpression of Snail had close relationship with the laryal squamous cell carcinoma, and it may be involved in the pre-cancerous cells to cancer cells into the regulation mechanism. Consistency were found between immunohistochemisty and in situ hybridization in detecting Snail, E-cadherin and VEGF. Snail, E-cadherin and VEGF mihgt take part in the occurrence, development, infiltration and metastasis of the larynal squamous cell carcinoma, which were important biological markers in the infiltration and metastasis. Co-detection of Snail, E-cadherin and VEGF might have important significance to evaluate the metastasis and prognosis of the tumors. The Snail could be as a new marker of potential therapeutic target in the laryanl squamous cell carcinoma.
