Expressions Of Twist And E-cadherin In Human Gastric Carcinoma And Clinical Pathological Significance

[Objective]To detect the expression of Twist and E-cadherin in the adjacent tissue of carcinoma and human gastric carcinoma tissue,and their clinical pathological correlations in human gastric carcinoma,Therefore we can more exactly decide the prognosis of gastric carcinoma in molecular level,and provide theory basis for the corresponding gene therapy method.[Methods] A total of 60 paraffin-embedded gasteric carcinoma and 20 adjacent tissue of cancer were studied by immunohistochemistry to evaluate the protein expression status of Twist and E-cadherinand.The relation was analyzed among the protein expression and age,stage,histological classification,cell differentiation and lymphatic metastasis.And detect the relationship of the two factors'expression.PBS instead of the frist Ab as a negative control and normal tissue as a positive control.[Results] The positive rate of Twist in gastric mucosa adjacent is 10%. The positive expression rate of Twist in gastric carcinoma was significantly enhanced,in this experiment Twist positive expression is 55.0%(33/60),the difference is significant(P<0.001).InⅠ,ⅡandⅢ,Ⅳstage gastric carcinoma,the positive expression rate of Twist was 20.0%(3/15),52.6% (10/19),65.4%(17/26),and there was a significant difference (P＜0.05).There was a significant difference between early and late stage gastric carcinoma in the positive expression rate of Twist.The positive expression rate of Twist was 61.5%(16/26) in case with lymph node metastasis, in contrast 38.5%(12/34)in cases without lymph node metastasis (P<0.05).There was a significant difference in E-cadherin expression between gastric carcinoma(28.3%) and adjacent tissue of cancer(90.0%) (P＜0.001).The nuclear or cytoplasm expression rate was 71.7%(43/60) regards as the cell membrane expression to reduce.In Well,moderately and poorly differentiated gastric carcinoma E-cadherin showd a reduced expression rate 60.0%(12/20), 65.0%(13/20),90.0%(18/20).There was a significant difference in E-cadherin expression among well,moderately and poorly,differentiated gasteric carcinoma(P=0.005). The expression of E-cadherin in poorly differentiated gasteric carcinoma was much lower than in well or moderately differentiated gasteric carcinoma.Reduced E-cadherin expression rate was 80.8%(21/26) in case with lymph node metastasis, in contrast 64.7%(22/34) in cases without lymph node metastasis (P=0.019)[Conclusion] The expression of Twist is associated with TNM stage and lymph node positive human gasteric carcinoma.The reduced membranous expression of E-cadherin is associated with poorly differentiated and lymph node positive human gasteric carcinoma.The expression of Twist is inversely correlated with nuclear expression rate of E-cadherin.
