Objective:Through the experimentation on animals, observe the changeof skin lesions on the guinea pigs skin before and after treatment,evaluate theeffect of Chinese herb combined with chitosan in treat of post-inflammatoryhyperpigmentation on the guinea pigs skin, provide reference for deeperfuture explore of the effective methods to treat and prevent the post-inflammatory hyperpigmentation.Methods:There are15brownish guinea pigs in this experiment totally,provided from the Experimental Animal Center of Hebei MedicalUniversity(Experimental animal production license number:1403134).Shaveoff the back of the brownish hair of guinea pigs,the exposed skin wererandomly divided into four separated regions.The carbon dioxide lattice laserirradiated guinea pigs dorsal skin,then the narrow band ultraviolet(wavelength311nm) irradiated.Established the animal model of post-inflammatory hyperpigmentation.The experimental group used Chinese herbalextract combined with chitosan biological membrane application,Chineseherbal extract was extracted by white poria cocos licorice and purslane,Chitosan is a natural moisturizer of skin(Product Standard No: YZB-Ji0033-2010), the control groups using the Q-switched Nd:YAG laser (wavelength1064nm),2%Hydroquinone Cream coated with and without any intervention.To observe the effect of fluence on the treatment of the brownish guinea pigsskin using different method. Through the VISIA image analysis results andmaterial tissue pathology after8weeks to observe the changes of pigment-ation. Analysis of different treatment methods on guinea pig pigmentationimprovement.After SPSS13.0statistical software analysis, P <0.05for thedifference was statistically significant. Results: In the process of this study,a total of15experimental animals,back every guinea pig from four separated regions,randomly divided into fourgroups,each group consists of15cases.Four groups of Absolute Scores ofP>0.05after statistical software analysis,no significant difference between thefour groups,the equilibrium can be compared.8weeks after intervention,record the changes respectively in Chinese medicine extract with chitosangroup,2%hydroquinone group,Q-switch Nd:YAG laser group and blankcontrol group of guinea pig skin brown spot of absolute scores.1Comparison the absolute scores of brown spot:8weeks after theintervention,guinea pigs four separated by regional changes in skin color.Between the four groups by analysis of variance,P<0.05,with significantdifference in statistically. Then,the experimental group compared with controlgroups were compared:Q-switched Nd:YAG laser group compared withChinese herbal extract combined with chitosan,P<0.05,with significantdifference in statistically;2%hydroquinone group compared with Chineseherbal extract combined with chitosan, P>0.05,no statistically significance,thetwo groups has the same treatment effect.And the Chinese herbal extractgroup has no stimulation,skin medication process does not appeared obviousredness and peeling phenomenon;The blank control group after8weeks,theabsolute value of guinea pig skin pigmentation brown spot no obviouslydecreased.2Comparison of histopathology: After three times exposure by311nmNB-UVB, the back skin of guinea pig form a uniform and stable pigmentat-ion.The basal layer of the epidermis showed a large amount of melaningranules,spinous layer visible melanin granules cap shaped distribution. After8weeks of treatment,Chinese herbal combined with chitosan group ofmelanin granules decreased significantly,the basal and spinous layers visiblein a few scattered melanin granules.2%Hydroquinone Cream group,thepigment particles are reduced,the basal and spinous layer is also a smallamount of melanin granules;Q-switch Nd:YAG laser group,the basal layer ofmelanin granules decreased slightly,spinous layer still visible melanin granules;The blank control groups pigment granules did not changesignificantly.Conclusion:The carbon dioxide lattice laser combined with311nm NB-UVB could successfully induce the model of post-inflammatory hyperpig-mentation in brownish guinea pigs.Chinese herbal extract combined withchitosan has a obvious improvement effect on post-inflammatory hyperpigm-entation of guinea pigs,it can decrease the absolute value of brown spot,shorten the duration time of post-inflammatory hyperpigmentation.The short-term effect of the Chinese herbal extract combined with chitosan is close tothe2%hydroquinone,and no obvious side effects.
