Objective:To compare the similarity of serum containing Buzhong Yiqi Decoction containing Radix Astragali and containing Radix Hedysari acting on spleen T lymphocytes and anti-aging of aging mice.Methods:1.120healthy kunming mice were randomly divided into3groups:Buzhong Yiqi Decoction containing Radix Hedysari、Buzhong Yiqi Decoction containing Radix Astragali and blank serum group, each group of40. Traditional Chinese medicine group were given4g crude drugs/(kg· d), blank serum group was given0.9%saline, quantity of three groups of mice were0.4ml/(20g· d), once per day,15d. The last1h after the treatment, eyes took blood, and to prepare the different concentrations of serum containing drug to culcure with aging mice spleen cell, through MTT method to determine the best drug-containing serum concentration and the best training time.2.The best drug-containing serum concentration of Buzhong Yiqi Decoction cultured with aging mice spleen lymphocytes to detect the proliferative capacity of T lymphocytes, the percentage of T-lymphocyte subsets and the level of IL-2in the culture supernatant.3.The best drug-containing serum concentration of Buzhong Yiqi Decoction cultured with aging mice spleen lymphocytes to observe intracellular SOD activity, content of the change on MDA and ROS.Results:1. MTT results showed that the spleen cell proliferation activity increased after decreased first with serum concentrations of Buzhong Yiqi Decoction containing Radix Hedysari and Buzhong Yiqi Decoction containing Radix Astragali increased and the extension of incubation time,40%of drug concentration with serum training48h stimulated proliferation best acting on spleen lymphocyte in aged mice when Buzhong Yiqi Decoction containing Radix Astragali and containing Radix Hedysari acting on spleen lymphocyte in aged mice.2. It could significantly increase T lymphocyte proliferation(P<0.05), the T lymphocyte count of CD28+in T lymphocyte subgroup(P<0.05), and the level of IL-2(P<0.01). Radix Hedysari was superb to Radix Astragali in Buzhong Yiqi Decoction in raising T lymphocyte proliferationand and IL-2level (P <0.05).3. At the same time, it also could significantly modulate aging mouse spleen lymphocyte SOD(P<0.01), MDA activity and intracellular reactive ROS level(P<0.05), there were no significant differences between Buzhong Yiqi Decoction containing Radix Hedysari and Radix Astragali(P>0.05).Conclusion:Buzhong Yiqi Decoction containing Radix Hedysari has a similar role in anti-Immunosenescence and anti-aging with containing Radix Astragali. Buzhong Yiqi Decoction containing Radix Hedysari has better effect in T cell proliferation and IL-2level, both have the replaceability.
