1. Comparative study on the interchange of Radix Astragali and Radix Hedyseri in Yiqiyangxue decoction and their effects on anti-oxidant injury, anti-immunosenescence and spleen lymphocyte proliferation in aging miceObjective:We substituted the same dose of Radix Hedyseri for Radix Astragali and intragastric administrated the mice once per day for14days to obtain drug serum. We used MTT method to determine the best drug serum concentration and the time of culturing and also evaluated the protective efficacy of anti-oxidant injury, anti-immunosenescence and drug induced lymphocyte proliferation.Method:Randomly divided120normal mice into three groups (containing Radix Hedyseri and Radix Astragali groups and normal saline group) and oral administrated them once per day for14days with different drug decoctions and normal saline respectively. After the final serve, orbital blood was obtained and centrifugalized to prepare the drug serum. Cervical dislocated the18-month aging mice and separated the spleen aseptically to prepare the spleen lymphocyte suspension. We used MTT method to evaluate the drug serum induced spleen lymphocyte proliferation in vitro and determine the best drug serum concentration and time of culturing. We co-cultured the drug serum and the spleen lymphocyte suspension to evaluate T-SOD, MDA, ROS, cultural supernatant IL-2level, and ConA induced lymphocyte proliferation.Results:Containing Radix Hedyseri and Radix Astragali in Yiqiyangxue decoction could both enhance the spleen lymphocyte proliferation of aging mice and containing Radix Hedyseri performed better. MTT results showed that40%of drug serum concentration co-cultured with lymphocyte suspension for72h performed the best efficacy in enhancing the lymphocyte proliferation. Both drug serums could increase T-SOD, IL-2and decrease MDA level and their efficacy were similar. But drug serum of Radix Hedyseri Yiqiyangxue performed better in the enhancement of IL-2and the reduction of ROS level.Conclusion:Both drug serums could enhance the spleen lymphocyte proliferation in vitro, and performed anti-oxidant injury effects. But Radix Hedyseri drug serum was superior in ConA-induced lymphocyte proliferation, increasing IL-2and reducing ROS level.2. Comparative study on alternation of Radix Hedyseri and Radix Astragali in Yiqiyangxue decoction and evaluation of their anti-oxidant injury and anti-immunosenescence effectsObjective:Alternated the same dose of Radix Hedyseri with Radix Astragali and oral administrated the18-month aging mice once per day for14days. After the final administration, cervical dislocated the aging mice and calculated the spleen and thymus index. Evaluate their anti-oxidant injury and anti-immunosenescence effects.Method:Randomly separated3018-month-old aging mice into three groups (Radix Hedyseri Yiqiyangxue group, Radix Astragali Yiqiyangxue group and Normal saline control group). Oral administrated the mice in each group once per day for14days with different drug decoctions and saline separately. After the final administration, cervical dislocated the mice then separated the spleen and thymus aseptically. Weighed the spleen and thymus accurately and calculated the spleen and thymus index then evaluated the serum T-SOD, MDA and IL-2levels.Results:Containing Radix Hedyseri and Radix Astragali in Yiqiyangxue decoction could both increase the serum T-SOD level and reduce MDA level. But no difference was found on the efficacy of both Radix Astragali and Radix Hedyseri. But Radix Hedyseri performed better in increasing spleen index and serum IL-2level.Conclusion:Both Yiqiyangxue prescriptions that containing Radix Hedyseri and Radix Astragali had the same effect against free radicals injuries. But Radix Hedyseri performed better in increasing serum IL-2level. This indicated that the two drugs were interchangeable on the efficacy against free radicals in vivo.
