1.Comparative the influence on immune function on the serum containing Fuqizhihan decoction of Radix Astragali with the serum containing Fuqizhihan decoction of Radix Hedyseri of Spleen Lymphocytes in SAMP8 mice.Objective:To compare the influence on immune function on the serum containing Fuqizhihan decoction of Radix Astragali with the serum containing Fuqizhihan decoction of Radix Hedyseri of Spleen Lymphocytes in SAMP8 mice.Method:Randomly divided 90 normal mice into three groups:Fuqizhihan Decoction containing Radix Astragali,Fuqizhihan Decoction containing Radix Hedyseri and blank serum group,each group of 30.Fuhong group and Fuhuang group were given 27g crude drugs/(kg.d),the thymosins group were intragastrically administrated the thymosins with 4.6 mg/kg,blank serum group was given 0.9%saline,0.4ml(20g.d),once per day.After the final serve,orbital blood was obtained and centrifugalized to prepare the drug serum.Cervical dislocated the SAMP8 mice and separated the spleen aseptically to prepare the spleen lymphocyte suspension.We used MTT method determine the optimum incubation time and serum concentrations,and detect the proliferation of the optimum serum on spleen T lymphocyte in SAMP8 mice.ConA induced lymphocyte proliferation.Flow cytometry to detect theCD3~+CD4~+?CD3~+CD8~+percentage of T lymphocyte subsets.ELISA detect the level of IL-2 in the culture supernate.qRT-PCR method test the mRNA expression in the PI3K.Results:MTT results showed that 40%of drug concentration with serum training 48 h stimulated proliferation best by acting on spleen lymphocyte in SAMP8 mice,and the medicated serums increased T lymphocyte proliferation and Radix Hedysari group was better than Radix Astragali group.It could increase T lymphocytic count of CD8~+and decrease T lymphocytic count of CD4~+in T lymphocytic subgroup(P<0.05).The serum both Radix Hedysari and Radix Astragali of Yupingfeng compound can increase the level of IL-2(P<0.05)and the expression of mRNA in the PI3K.Radix Hedysari was superior to Radix Astragali on the expression of mRNA in the PI3K.Conclusion:Fuqizhihan compound containing Radix Hedysari and Radix Astragali all can modulate immune,It can play a positive role in regulating the immune function in order to slow down the aging.2.Comparative study of Radix Hedyseri and Radix Astragali alternative Fuqizhihan decoction in influence on immune function of Spleen Lymphocytes in SAMP8 mice.Objective:To compare the effects of Fuqizhihan decoction contain Radix Astragali or Radix Hedyseri on the anti immunosenescence function of Spleen Lymphocytes in SAMP8 mice.Method:Ten Kunming mice were taken as normal group.40 SAMP8 mice were randomly divided as saline group,thymosins group,Fuqizhihan Radix Hedyseri group and Fuqizhihan Radix Astragali group.The mice in the normal group and the saline group were intragastrically administrated with the same volume of distilled water,in the thymosins group were intragastrically administrated the thymosins with 4.6 mg/kg,once a day for 14 times.Drawing blood through ophthalmectomy after 2 hours of the last irrigation,while taking the spleen.To detect the level of IFN-?in the serum of the mice in each group by ELISA.To detect the effect of Fuqizhihan decoction on T cells differentiation with Flow cytometry.The Fuqizhihan Decoction effection of protein expression level of PI3K in T lymphocyte were investigated by using Western-blot,qRT-PCR method test the mRNA expression in the PI3K.Results:Compared with the normal group,The Fuqizhihan Decoction containing Radix Hedysari or Radix Astragali increased production of IFN-?in serum of SAMP8 mice(P<0.05),but the thymosins group contained higher than the decoctions groups.The expression of CD28~+was improved and the expression of CD152~+was decreased(P<0.05).Both Fuqizhihan Decoction containing Radix Hedysari and Fuqizhihan Decoction containing Radix Astragali increased the relative expression of PI3K protain and PI3K mRNA in SAMP8 mice(P<0.05).Conclusion:Both Fuqizhihan compound containing Radix Hedysari and Radix Astragali through regulating the immune function of Spleen Lymphocytes in SAMP8 mice to retard the aging process.and Fuqizhihan compound containing Radix Hedysari and Radix Astragali has the similar role.
